[Objectives]miRNAs play an important role in the proliferation and differentiation of different myoblasts.This study was conducted to elucidate the complex genetic mechanisms that affect the meat production performanc...[Objectives]miRNAs play an important role in the proliferation and differentiation of different myoblasts.This study was conducted to elucidate the complex genetic mechanisms that affect the meat production performance of Sichuan white rabbits and reveal the regulatory role of miRNAs in their muscle growth and meat quality formation.[Methods]Three constructed skeletal muscle libraries of Sichuan white rabbits aged six months were sequenced by the solexa technology to identify known miRNAs,predict new miRNAs and construct an expression profile of muscle miRNAs.[Results]A total of 511 known miRNAs and 42 miRNAs were detected in 34089472 pure sequences,and the proportion of miRNAs with a length of 22 nt was the highest.The number of known miRNA sequences accounted for 71.38%of pure sequences,which was much higher than the proportion of other types of RNAs.The proportion of sequences from exons was 0.38%,indicating a low degree of mRNA degradation in the samples.Base U had the highest proportion at the first position,and the bases with the highest proportions at positions 8 and 10 were U and A,respectively.Muscle-specific miRNAs(miR-1,miR-133,and miR-206)ranked in the top 10 in terms of expression level.The number and expression levels of new miRNAs were lower than those of known miRNAs.The length distribution,base bias at different positions and expression profile characteristics of miRNAs might be related to the biological function of miRNAs in regulating muscle proliferation and differentiation and the action mechanisms with target genes.[Conclusions]The identification and expression of miRNAs in muscle tissues of Sichuan white rabbits will help to understand the complex molecular mechanisms of meat production performance and provide a theoretical basis for the functional research of miRNAs in meat rabbits.展开更多
DNA methyltransferases(Dnmts)comprise a family of proteins which involved in the establishment and maintenance of DNA methylation patterns.In pig,the molecular characterization and tissue expression profile of Dnmt ge...DNA methyltransferases(Dnmts)comprise a family of proteins which involved in the establishment and maintenance of DNA methylation patterns.In pig,the molecular characterization and tissue expression profile of Dnmt gene family are not clear.To solve this problem,reverse transcriptase PCR and rapid amplification of c DNA ends were used to clone the sequences of the porcine Dnmt2 and Dnmt3b genes.Furthermore,the m RNA expression profiles of Dnmt1,Dnmt2,Dnmt3a and Dnmt3b genes from 54 adult tissues and 2 entire fetuses of Rongchang pig were analyzed by quantitative real-time PCR(q RT-PCR).As a result,the lengths of porcine Dnmt2 and Dnmt3b gene c DNAs were 1 227 and 2 559 bp with cytosine-C5specific DNA methylase domain,respectively.The four Dnmt genes were highly expressed in longissimus dorsi muscle(P<0.01).Dnmt1 is highly expressed in heart(P<0.01)and Dnmt 2 shows its preference in liver and seminal vesicle tissue(P<0.01).Dnmt3a and Dnmt3b are highly expressed in the two fetus stages(P<0.01).All these results suggested that each gene has its specific expression profile,and deeper study is required to dig more details between the methylation level and Dnmt family m RNA expressions in different tissues.展开更多

The results showed that the respiration,body temperature and heart rate of Tibetan sheep at 3- 24 months of age were( 24. 60 ± 6. 33)-( 50. 30 ± 21. 18) times / min,( 39. 14 ± 0. 28) ℃-( 39. 64 ± 0. 17) ℃ and( 91. 00 ± 13. 27)-( 112. 00 ± 8. 69) times/min,respectively. The blood physiological indexes of Tibetan sheep including white blood cell count( WBC),hematocrit( HCT) and red blood cell distribution width coefficient of variation( RDW- CV) at 3,6,15 and 18 months of age were relatively lower than those at 9,12,21 and 24 months of age. The blood physiological indexes including red blood cell count( RBC),mean corpuscular capacity( MCV),mean corpuscular hemoglobin concentration( MCHC) and mean corpuscular hemoglobin( MCH) at 3,6,15 and 18 months of age were relatively higher than those at 9,12,21 and 24 months of age,while hemoglobin( HGB) and platelet count( PLT) did not vary with months of age. For blood biochemical indexes,alanine aminotransferase( ALT),albumin( ALB),globulin( GLO) and total cholesterol( CHOL) reached the maximum values at 21 months of age; alkaline phosphatase( ALKP),lactate dehydrogenase( LDH),blood glucose( GLU) and pseudocholinesterase( PCHE) reached the maximum values at 3 months of age;and total calcium( CA),total protein( TP),aspartate aminotransferase( AST) reached the maximum values at 9,18 and 24 months of age,respectively.展开更多

This study was to investigate the expression pattern of IGF-I gene and correlation between its SNPs and growth traits in sheep.The expression patterns of IGF-I gene in brain,muscle,skin,liver and heart of Liangshan se...This study was to investigate the expression pattern of IGF-I gene and correlation between its SNPs and growth traits in sheep.The expression patterns of IGF-I gene in brain,muscle,skin,liver and heart of Liangshan semi-wool sheep were detected at six growth stages(15,60,105,195,240 days) by employing qRT-PCR approach,and the correlation between SNPs of IGF-I gene and growth traits via SSCP method.The first inflection point of IGF-I gene expression was observed at day 105 in all five tissues.From 15-105 days IGF-I gene gave a decline-ascending trend in muscle,brain and liver,while it did not vary significantly during day 15-day 60 and was up-regulated in heart and liver at day 105.Whenafter,the expression level was down-regulated in liver,while that in muscle,heart,brain and skin assumed the second inflection point.The expression levels of IGF-I gene in brain and skin at day 195 were significantly higher than that at day 150 and day 240.These suggest the synchronous expression of IGF-I gene in all five tissues.Two SNPs located at the leader region and exon 3 of IGF-I gene were middle or low polymorphic.The SNP locus detected with primer P-1 was significantly correlated with birth weight of Liangshan semi-wool sheep(P <0.05),while that detected with primer P-2 was significantly correlated with weaning weight and the weaning daily gain(P < 0.05).The expression of IGF-I in all tissues had the similar developmental patterns,and strong correlation of the SNPs of IGF-I gene was confirmed with the eary growth traits to provide theoretical basis for the growth regulation and applying to assisted selective breeding with the SNPs in liangshan semi-wool sheep.展开更多

The results showed that t...In this study,using Liangshan semi-fine wool sheep as an experimental material,full-length CDS sequence of IGFBP-1 gene was cloned with RT-PCR method and analyzed with bioinformatics methods. The results showed that the full-length CDS sequence of IGFBP-1 gene in Liangshan semi-fine wool sheep was792 bp in length,encoding 263 amino acids. The CDS sequence shared 97%,76% and 74% homology with bovine,human and rat,respectively; the amino acid sequence shared 97%,69% and 71%,respectively. The Gen Bank accession number was FJ589639. 1. The amino acid molecular weight of IGFBP-1 was 27. 8k D,and the theoretical isoelectric point( p I) was 5. 99. The result of phylogenetic analysis showed that IGFBP-1 gene in Liangshan semi-fine wool sheep exhibited close phylogenetic relationships with bovine,goat and other mammals,and distant phylogenetic relationships with chicken and fish. IGFBP-1 gene had obvious hydrophobic and hydrophilic regions,harboring one signal peptide,one transmembrane region,16 phosphorylation sites,six N-glycosylation sites and eight O-glycosylation sites. The result of secondary structure analysis showed that the random coil,α-helix and β-sheet regions accounted for 61. 98%,24. 33% and 13. 69%,respectively. The result of tertiary structure analysis showed that IGFBP-1 harbors an IGFBP_N domain and a thyroglobulin type-I domain. The results showed that the body temperature,respiration and heart rate of Tibetan sheep at four seasons were 39. 18- 39. 60 ℃,27. 20- 49. 04 times / min,91. 87- 106. 25 times / min,respectively. The indexes of WBC,MCV,MCH and MCHC in autumn and winter were significantly higher than those in spring and summer( P < 0. 01),while the indexes of RBC,HCT and RDW-CV in spring and summer were significantly higher than those in autumn and winter( P < 0. 01). The PLT did not vary greatly in spring,summer or autumn,but were all extremely higher than that in winter( P< 0. 01). The HGB maintained stable in the four seasons. The indexes of TP,ALB and GLO in summer and autumn were extremely higher than those in winter and spring( P < 0. 01),while the indexes of ALP and PCHE in summer and autumn were extremely lower than those in winter and spring( P < 0. 01). Objective:Flavonoids are the bioactive compounds in safflower(Carthamus tinctorius),in which chalcone synthase(CHS)is the first limiting enzyme.However,it is unclear that which chalcone synthase genes(CHSs)are partici...Objective:Flavonoids are the bioactive compounds in safflower(Carthamus tinctorius),in which chalcone synthase(CHS)is the first limiting enzyme.However,it is unclear that which chalcone synthase genes(CHSs)are participated in flavonoids biosynthesis in C.tinctorius.In this study,the CHSs in the molecular characterization and enzyme activities were investigated.Methods:Putative chalcone biosynthase genes were screened by the full-length transcriptome sequences data in C.tinctorius.Chalcone biosynthase genes in C.tinctorius(CtCHSs)were cloned from cDNA of flowers of C.tinctorius.The cloned gene sequences were analyzed by bioinformatics,and their expression patterns were analyzed by real-time PCR(RT-PCR).The protein of CtCHS in the development of flowers was detected by polyclonal antibody Western blot.A recombinant vector of CtCHS was constructed.The CtCHS recombinant protein was induced and purified to detect the enzyme reaction(catalyzing the reaction of p-coumaryl-CoA and malonyl-CoA to produce naringin chalcone).The reaction product was detected by HPLC and LC-MS.Results:Two full-length CtCHS genes were successfully cloned from the flowers of safflower(CtCHS1 and CtCHS3),with gene lengths of 1525 bp and 1358 bp,respectively.RT-PCR analysis showed that both genes were highly expressed in the flowers,but the expression of CtCHS1 was higher than that of CtCHS3 at each developmental stage of the flowers.WB analysis showed that only CtCHS1 protein could be detected at each developmental stage of the flowers.HPLC and LC-MS analyses showed that CtCHS1 could catalyze the conversion of p-coumaryl-CoA and malonyl-CoA substrates to naringin chalcone.Conclusion:CtCHS1 is involved in the biosynthesis of naringin chalcone in safflower.展开更多

文摘In order to compare the physiological and biochemical indexes of different sheep populations in plateau area,we measured the physiological and biochemical indexes of White Suffolk sheep,White-tibetan sheep and Tibetan sheep in four seasons.The body temperature,respiration and heart rate of White Suffolk sheep,White-tibetan sheep and Tibetan sheep throughout the year were(38.96±0.23)-(40.80±0.74)℃,(27.20±7.73)-(74.56±10.91)times/min,and(83.10±2.42)-(106.25±7.07)times/min,respectively.The main blood physiological indexes of White-tibetan sheep were between those of Suffolk sheep and Tibetan sheep or close to that of Tibetan sheep,the indexes of WBC in summer,HGB and RDW-CV in autumn,RDW-CV in winter were close to that of White Suffolk sheep.The main blood biochemical indexes of White-tibetan sheep were between that of White Suffolk sheep and Tibetan sheep or close to that of Tibetan sheep,but the indexes of ALB in spring,AST in summer,ALP and CHOL in autumn,and ALP in winter were close to that of White Suffolk sheep.It revealed that the adaptability of White-tibetan sheep was prior to White Suffolk sheep,and tended to Tibetan sheep in the plateau environment.

文摘The change in lactation of Maiwa yak was studied based on the data of locating Maiwa yak in warm seasons during 2003- 2009. The results showed that the milk yield was increasing gradually during days 30- 70 after calving,reached the peak at days 70- 110,and decreased gradually to no milking whenafter. The milk yield of Maiwa yak was greatly influenced by lactation age rather than the yearly effect.

文摘This study was to investigate the expression pattern of IGF-I gene and correlation between its SNPs and growth traits in sheep.The expression patterns of IGF-I gene in brain,muscle,skin,liver and heart of Liangshan semi-wool sheep were detected at six growth stages(15,60,105,195,240 days) by employing qRT-PCR approach,and the correlation between SNPs of IGF-I gene and growth traits via SSCP method.The first inflection point of IGF-I gene expression was observed at day 105 in all five tissues.From 15-105 days IGF-I gene gave a decline-ascending trend in muscle,brain and liver,while it did not vary significantly during day 15-day 60 and was up-regulated in heart and liver at day 105.Whenafter,the expression level was down-regulated in liver,while that in muscle,heart,brain and skin assumed the second inflection point.The expression levels of IGF-I gene in brain and skin at day 195 were significantly higher than that at day 150 and day 240.These suggest the synchronous expression of IGF-I gene in all five tissues.Two SNPs located at the leader region and exon 3 of IGF-I gene were middle or low polymorphic.The SNP locus detected with primer P-1 was significantly correlated with birth weight of Liangshan semi-wool sheep(P <0.05),while that detected with primer P-2 was significantly correlated with weaning weight and the weaning daily gain(P < 0.05).The expression of IGF-I in all tissues had the similar developmental patterns,and strong correlation of the SNPs of IGF-I gene was confirmed with the eary growth traits to provide theoretical basis for the growth regulation and applying to assisted selective breeding with the SNPs in liangshan semi-wool sheep.

文摘To understand the exact changes in body size and body weight of Jinchuan female yaks within one year,the body size and body weight of 30 Jinchuan female yaks were measured from July 2015 to July 2016. The results showed that the body weight of Jinchuan female yak declined continuously since October 2015,and the weight loss in November 2015 was the largest. The body weight started to increase in May 2016,and the body weight increased significantly in June and July of 2016.

文摘In this study,using Liangshan semi-fine wool sheep as an experimental material,full-length CDS sequence of IGFBP-1 gene was cloned with RT-PCR method and analyzed with bioinformatics methods. The results showed that the full-length CDS sequence of IGFBP-1 gene in Liangshan semi-fine wool sheep was792 bp in length,encoding 263 amino acids. The CDS sequence shared 97%,76% and 74% homology with bovine,human and rat,respectively; the amino acid sequence shared 97%,69% and 71%,respectively. The Gen Bank accession number was FJ589639. 1. The amino acid molecular weight of IGFBP-1 was 27. 8k D,and the theoretical isoelectric point( p I) was 5. 99. The result of phylogenetic analysis showed that IGFBP-1 gene in Liangshan semi-fine wool sheep exhibited close phylogenetic relationships with bovine,goat and other mammals,and distant phylogenetic relationships with chicken and fish. IGFBP-1 gene had obvious hydrophobic and hydrophilic regions,harboring one signal peptide,one transmembrane region,16 phosphorylation sites,six N-glycosylation sites and eight O-glycosylation sites. The result of secondary structure analysis showed that the random coil,α-helix and β-sheet regions accounted for 61. 98%,24. 33% and 13. 69%,respectively. The result of tertiary structure analysis showed that IGFBP-1 harbors an IGFBP_N domain and a thyroglobulin type-I domain. This study laid a solid foundation for further investigating the function of IGFBP-1 gene in sheep.

The result of secondary structure analysis showed that the random coil,α-helix and β-sheet regions accounted for 64. 89%,19. 86% and 15. 25%,respectively. The result of tertiary structure analysis showed that IGFBP-7 harbors an IGFBP_N domain and an Ig-like domain. This study provided scientific basis for further investigating the function of IGFBP-7 gene in sheep.

文摘To investigate the seasonal changes in physiological and biochemical indexes of Tibetan sheep in Hongyuan area,we measured the physiological and biochemical indexes of Tibetan sheep at four different seasons. The results showed that the body temperature,respiration and heart rate of Tibetan sheep at four seasons were 39. 18- 39. 60 ℃,27. 20- 49. 04 times / min,91. 87- 106. 25 times / min,respectively. The indexes of WBC,MCV,MCH and MCHC in autumn and winter were significantly higher than those in spring and summer( P < 0. 01),while the indexes of RBC,HCT and RDW-CV in spring and summer were significantly higher than those in autumn and winter( P < 0. 01). The PLT did not vary greatly in spring,summer or autumn,but were all extremely higher than that in winter( P< 0. 01). The HGB maintained stable in the four seasons. The indexes of TP,ALB and GLO in summer and autumn were extremely higher than those in winter and spring( P < 0. 01),while the indexes of ALP and PCHE in summer and autumn were extremely lower than those in winter and spring( P < 0. 01). The indexes of AST,ALT,LDH,GLU,CHOL and CA maintained stable in all seasons.

文摘Objective:Flavonoids are the bioactive compounds in safflower(Carthamus tinctorius),in which chalcone synthase(CHS)is the first limiting enzyme.However,it is unclear that which chalcone synthase genes(CHSs)are participated in flavonoids biosynthesis in C.tinctorius.In this study,the CHSs in the molecular characterization and enzyme activities were investigated.Methods:Putative chalcone biosynthase genes were screened by the full-length transcriptome sequences data in C.tinctorius.Chalcone biosynthase genes in C.tinctorius(CtCHSs)were cloned from cDNA of flowers of C.tinctorius.The cloned gene sequences were analyzed by bioinformatics,and their expression patterns were analyzed by real-time PCR(RT-PCR).The protein of CtCHS in the development of flowers was detected by polyclonal antibody Western blot.A recombinant vector of CtCHS was constructed.The CtCHS recombinant protein was induced and purified to detect the enzyme reaction(catalyzing the reaction of p-coumaryl-CoA and malonyl-CoA to produce naringin chalcone).The reaction product was detected by HPLC and LC-MS.Results:Two full-length CtCHS genes were successfully cloned from the flowers of safflower(CtCHS1 and CtCHS3),with gene lengths of 1525 bp and 1358 bp,respectively.RT-PCR analysis showed that both genes were highly expressed in the flowers,but the expression of CtCHS1 was higher than that of CtCHS3 at each developmental stage of the flowers.WB analysis showed that only CtCHS1 protein could be detected at each developmental stage of the flowers.HPLC and LC-MS analyses showed that CtCHS1 could catalyze the conversion of p-coumaryl-CoA and malonyl-CoA substrates to naringin chalcone.Conclusion:CtCHS1 is involved in the biosynthesis of naringin chalcone in safflower.
