The present study was designed to explore the mechanisms underlying improved motor function via transcranial magnetic stimulation and rehabilitation training following cerebral infarction. Results showed that rehabilitation training or transcranial magnetic stimulation alone reduced neurological impairment in rats following cerebral infarction, as well as significantly increased synaptic curvatures and post-synaptic density in the non-injured cerebral hemisphere sensorimotor cortex and narrowed the synapse cleft width. In addition, the percentage of perforated synapses increased. The combination of transcranial magnetic stimulation and rehabilitation resulted in significantly increased total dendritic length, dendritic branching points, and dendritic density in layer V pyramidal cells of the non-injured cerebral hemisphere motor cortex. These results demonstrated that transcranial magnetic stimulation and rehabilitation training altered structural parameters of pyramidal cell dendrites and synapses in the non-injured cerebral hemisphere sensorimotor cortex, thereby improving the ability to compensate for neurological functions in rats following cerebral infarction.展开更多

Stem cell transplantation has brought new hope for the treatment of neurological diseases.The key to stem cell therapy lies in inducing the specific differentiation of stem cells into nerve cells.Because the different...Stem cell transplantation has brought new hope for the treatment of neurological diseases.The key to stem cell therapy lies in inducing the specific differentiation of stem cells into nerve cells.Because the differentiation of stem cells in vitro and in vivo is affected by multiple factors,the final differentiation outcome is strongly associated with the microenvironment in which the stem cells are located.Accordingly,the optimal microenvironment for inducing stem cell differentiation is a hot topic.EGb761 is extracted from the leaves of the Ginkgo biloba tree.It is used worldwide and is becoming one of the focuses of stem cell research.Studies have shown that EGb761 can antagonize oxygen free radicals,stabilize cell membranes,promote neurogenesis and synaptogenesis,increase the level of brain-derived neurotrophic factors,and replicate the environment required during the differentiation of stem cells into nerve cells.This offers the possibility of using EGb761 to induce the differentiation of stem cells,facilitating stem cell transplantation.To provide a comprehensive reference for the future application of EGb761 in stem cell therapy,we reviewed studies investigating the influence of EGb761 on stem cells.These started with the composition and neuropharmacology of EGb761,and eventually led to the finding that EGb761 and some of its important components play important roles in the differentiation of stem cells and the protection of a beneficial microenvironment for stem cell transplantation.展开更多

METHODS: A total of 36 unselected, frozen samples of cholangiocarcinoma were collected, p53 gene status(exon 5-8) and P53 protein were examined by automated sequencing and immunohistochemical staining, combined with the clinical parameters of patients. RESULTS: P53 gene mutations were found in 22 of 36 (61.1%) patients. Nineteen of 36 (52.8%) patients were positive for P53 protein expression. There were significant differences in extent of differentiation and invasion between the positive and negative expression of P53 protein. However, there were no significant differences in pathologic parameters between the mutations and non-mutations. CONCLUSION: The alterations of the P53 gene evaluated by DNA sequence analysis is relatively accurate. Expression of P53 protein could not act as an independent index to estimate the prognosis of cholangiocarcinoma.展开更多

The InsR expression and TP in skeleton muscle tissue were determined by Western blotting and immunoprecipitation in women with GDM (GDM group, n=22), normal pregnant women (normal pregnancy group, n=22) and normal non-pregnant women (normal non-pregnant group, n=13). Fasting plasma glucose (FPG) and fasting insulin (FINS) were measured by oxidase assay and immunoradioassay. The results showed that the levels of FPG (5.61±0.78 mmol/L), FINS (15.42±5.13 mU/L) and Ho- meostasis model assessment-IR (HOMA-IR) (1.21±0.52) in GDM group were significantly higher than those in normal pregnancy group (4.43±0.46 mmol/L, 10.56±3.07 mU/L and 0.80±0.31 respectively) (P〈0.01). The levels of FINS and HOMA-IR in normal pregnancy group were significantly higher than those in normal non-pregnant group (7.56±2.31 mU/L and 0.47±0.26 respectively) (P〈0.01). There was no significant difference in the InsR expression level among the three groups (P〉0.05). TP of InsR with insulin stimulation was significantly decreased in GDM group (0.20±0.05) as compared with normal pregnancy group (0.26±0.06) (P〈0.01). TP of InsR with insulin stimulation in normal pregnancy group was lower than that in normal non-pregnant group (0.31±0.06) (P〈0.01). TP of InsR with insulin stimu- lation was negatively related with HOMA-IR in GDM group (r=-0.525, P〈0.01). There was no correlation between the protein expression of InsR and HOMA-IR in GDM group (r=-0.236, P〉0.05). It was suggested that there is no significant correlation between the protein expression of InsR in skeletal muscle and IR in GDM, but changes in TP of InsR are associated with IR in GDM.展开更多
Objective Iodothyronine deiodinases(DIOs)are important selenoproteins that play a key role in the bone and joint diseases.Osteoarthritis(OA)is the most prevalent joint disease especially in elders.This bioinformatic a...Objective Iodothyronine deiodinases(DIOs)are important selenoproteins that play a key role in the bone and joint diseases.Osteoarthritis(OA)is the most prevalent joint disease especially in elders.This bioinformatic analysis was performed to explore the role of DIOs in OA pathogenesis.Methods The biological functions of selenoprotein DIOs were analyzed by bioinformatic techniques,mcluding GenCLip 3.0,Database for Annotation,Visualization and Integrated Discovery(DAVID),STRING,Cytoscape,and Network Analyst.The expression of DIOs in the healthy individuals and OA patients was determined by mining OA-related microarray data in the gene expression omnibus(GEO)database of National Center for Biotechnology Information and performing a Meta-analysis of the data with Review Manager 5.3.Results Cluster analysis revealed that the function of the DIOs was associated with thyroid hormone receptor and iodothyronine;GO analysis showed that DIOs were mainly involved in biological processes,such as ethanol metabolism and phenol-containing compound metabolism and primarily involved in the cytochrome P450 metabolism of exogenous organisms and thyroid hormone signaling;SULT1A1 was the core node of the PPI network;miRNAs and thyroid hormones had some iterations with DIO1 and DI02;Meta-analysis showed that DIO3 expression was significantly up-regulated in OA patients(SMD=0.31,95%CI:0.03,0.59,P=0.03).Conclusions The main biological functions of DIOs were closely associated with the regulation of thyroid hormone.And the up-regulated expression of DIO3 may have crucial impact on the occurrence of OA.展开更多

Intestinal pseudo-obstruction was diagnosed by abdominal radiography. Distention of the right abdominal wall was still apparent after one month. In this report, we found that recovery from abdominal distention caused by herpes zoster is difficult and may require surgical intervention.展开更多
Endometrial cancer remains to be a major type of malignancy in threatening female life.Molecular insights in advancing our understanding of endometrial tumorigenesis are much needed.We here report that a less-studied ...Endometrial cancer remains to be a major type of malignancy in threatening female life.Molecular insights in advancing our understanding of endometrial tumorigenesis are much needed.We here report that a less-studied protein Dihydropyrimidinase like 3(DPYSL3)is a potent tumor suppressor.DPYSL3 is uniquely regulated by wild type p53(wtp53),and its expression is at the highest level when cells carry wtp53 and are exposed to hypoxia.We reveal that wtp53 can bind DPYSL3 promoter to enhance DPYSL3 expression and in turn,the elevated DPYSL3 can restrain cancer cell proliferation and invasion in vitro and in vivo.Importantly,we observe that DPYSL3 can interfere with MAP kinase pathway,supported by a substantially reduced level of phosphorylated ERK in cells with high expression of DPYSL3.Furthermore,we identify the specific region of DPYSL3 that is responsible for its interaction with MEK and a subsequently reduced activity of ERK.In combination of molecular docking and mutagenesis analysis,we validated that the therapeutic implication of 17 A.A.s of DPYSL3,which can reduce the activity of the MAPK pathway and inhibit endometrial tumor cell growth in vitro and in vivo.Therefore,our study not only demonstrates in-depth understanding of human tumorigenesis,especially endometrial tumor,but also only provides a therapeutic potential to develop an effective tool to fight against human malignancy.展开更多

[Results] It searched 3 articles related to clinical application and 111 cases of patients. BACKGROUND Gastrointestinal stromal tumor(GIST)is a common neoplasm with high rates of recurrence and metastasis,and its therapeutic efficacy is still not ideal.There is an unmet need to find new molecular therapeutic...BACKGROUND Gastrointestinal stromal tumor(GIST)is a common neoplasm with high rates of recurrence and metastasis,and its therapeutic efficacy is still not ideal.There is an unmet need to find new molecular therapeutic targets for GIST.TATA-boxbinding protein-associated factor 15(TAF15)contributes to the progress of various tumors,while the role and molecular mechanism of TAF15 in GIST progression are still unknown.AIM To explore new molecular therapeutic targets for GIST and understand the biological role and underlying mechanisms of TAF15 in GIST progression.METHODS Proteomic analysis was performed to explore the differentially expressed proteins in GIST.Western blotting and immunohistochemical analysis were used to verify the expression level of TAF15 in GIST tissues and cell lines.Cell counting kit-8,colony formation,wound-healing and transwell assay were executed to detect the ability of TAF15 on cell proliferation,migration and invasion.A xenograft mouse model was applied to explore the role of TAF15 in the progression of GIST.Western blotting was used to detect the phosphorylation level and total level of RAF1,MEK and ERK1/2.RESULTS A total of 1669 proteins were identified as differentially expressed proteins with 762 upregulated and 907 downregulated in GIST.TAF15 was selected for the further study because of its important role in cell proliferation and migration.TAF15 was significantly over expressed in GIST tissues and cell lines.Overexpression of TAF15 was associated with larger tumor size and higher risk stage of GIST.TAF15 knockdown significantly inhibited the cell proliferation and migration of GIST in vitro and suppressed tumor growth in vivo.Moreover,the inhibition of TAF15 expression significantly decreased the phosphorylation level of RAF1,MEK and ERK1/2 in GIST cells and xenograft tissues,while the total RAF1,MEK and ERK1/2 had no significant change.CONCLUSION TAF15 is over expressed in GIST tissues and cell lines.Overexpression of TAF15 was associated with a poor prognosis of GIST patients.TAF15 promotes cell proliferation and migration in GIST via the activation of the RAF1/MEK/ERK signaling pathway.Thus,TAF15 is expected to be a novel latent molecular biomarker or therapeutic target of GIST.展开更多

Gastroduodenal endoscopic examination revealed an oval-shaped submucosal tumor at the prepyloric area on the posterior wall of the stomach. A degenerated gastrointestinal stromal tumor was suspected. Distal gastrectomy was performed and a histological diagnosis of heterotopic pancreas (HPs) was confirmed. The patient had an uneventful postoperative course and was discharged 7 d a^er operation. The patient remains healthy and symptom-free in the follow-up of 6 mo. This is a report of a case of gastric outlet obstruction resulting from pancreatic heterotopia in the gastric antrum in an adult woman.展开更多
AIM To evaluate the efficacy of main portal vein stents combined with iodine-125(^(125)Ⅰ) to treat main portal vein tumor thrombus.METHODS From January 1, 2010 to January 1, 2015, 111 patients were diagnosed with liv...AIM To evaluate the efficacy of main portal vein stents combined with iodine-125(^(125)Ⅰ) to treat main portal vein tumor thrombus.METHODS From January 1, 2010 to January 1, 2015, 111 patients were diagnosed with liver cancer combined with main portal vein tumor thrombus. They were non-randomly assigned to undergo treatment with transarterial chemoembolization(TACE)/transarterial embolization(TAE) + portal vein stents combined with ^(125)Ⅰ implantation(Group A) and TACE/TAE + portal vein stents only(Group B). After the operation, scheduled follow-up was performed at 6, 12 and 24 mo. The recorded information included clinical manifestations, survival rate, and stent restenosis rate. Kaplan–Meier curves, log-rank test and Cox regression were used for data analyses. RESULTS From January 1, 2010 to January 1, 2015, 54 and 57 patients were allocated to Groups A and B, respectively. The survival rates at 6, 12 and 24 mo were 85.2%, 42.6% and 22.2% in Group A and 50.9%, 10.5% and 0% in Group B. The differences were significant [log rank P < 0.05, hazard ratio(HR): 0.37, 95%CI: 0.24-0.56]. The rates of stent restenosis were 18.5%, 55.6% and 83.3% in Group A and 43.9%, 82.5% and 96.5% in Group B. The differences were significant(log rank P < 0.05, HR: 0.42, 95%CI: 0.27-0.63). Cox regression identified that treatment was the only factor affecting survival rate in this study.CONCLUSION Main portal vein stents combined with ^(125)Ⅰ can significantly improve survival rate and reduce the rate of stent restenosis.展开更多
Peripheral nerve injuries occur as the result of sudden trauma and lead to reduced quality of life.The peripheral nervous system has an inherent capability to regenerate axons.However,peripheral nerve regeneration fol...Peripheral nerve injuries occur as the result of sudden trauma and lead to reduced quality of life.The peripheral nervous system has an inherent capability to regenerate axons.However,peripheral nerve regeneration following injury is generally slow and incomplete that results in poor functional outcomes such as muscle atrophy.Although conventional surgical procedures for peripheral nerve injuries present many benefits,there are still several limitations including scarring,difficult accessibility to donor nerve,neuroma formation and a need to sacrifice the autologous nerve.For many years,other therapeutic approaches for peripheral nerve injuries have been explored,the most notable being the replacement of Schwann cells,the glial cells responsible for clearing out debris from the site of injury.Introducing cultured Schwann cells to the injured sites showed great benefits in promoting axonal regeneration and functional recovery.However,there are limited sources of Schwann cells for extraction and difficulties in culturing Schwann cells in vitro.Therefore,novel therapeutic avenues that offer maximum benefits for the treatment of peripheral nerve injuries should be investigated.This review focused on strategies using mesenchymal stem cells to promote peripheral nerve regeneration including exosomes of mesenchymal stem cells,nerve engineering using the nerve guidance conduits containing mesenchymal stem cells,and genetically engineered mesenchymal stem cells.We present the current progress of mesenchymal stem cell treatment of peripheral nerve injuries.展开更多
Objective:Although T-cell immunoglobulin and mucin-domain containing molecule-3(Tim-3)has been recognized as a promising target for cancer immunotherapy,its exact role in breast cancer has not been fully elucidated.Me...Objective:Although T-cell immunoglobulin and mucin-domain containing molecule-3(Tim-3)has been recognized as a promising target for cancer immunotherapy,its exact role in breast cancer has not been fully elucidated.Methods:Tim-3 gene expression in breast cancer and its prognostic significance were analyzed.Associated mechanisms were then explored in vitro by establishing Tim-3-overexpressing breast cancer cells.Results:In a pooled analysis of The Cancer Genome Atlas(TCGA)database,Tim-3 gene expression levels were significantly higher(P<0.001)in breast cancer tissue,compared with normal tissues.Tim-3 was a prognosis indicator in breast cancer patients[relapse-free survival(RFS),P=0.004;overall survival(OS),P=0.099].Tim-3 overexpression in Tim-3 low breast cancer cells promoted aggressiveness of breast cancer cells,as evidenced by enhanced proliferation,migration,invasion,tight junction deterioration and tumor-associated tubal formation.Tim-3 also enhanced cellular resistance to paclitaxel.Furthermore,Tim-3 exerted its function by activating the NF-κB/STAT3 signalling pathway and by regulating gene expression[cyclin D1(CCND1),C-Myc,matrix metalloproteinase-1(MMP1),TWIST,vascular endothelial growth factor(VEGF)upregulation,concomitant with Ecadherin downregulation).Lastly,Tim-3 downregulated tight junction-associated molecules zona occludens(ZO)-2,ZO-1 and occludin,which may further facilitate tumor progression.Conclusions:Tim-3 plays an oncogenic role in breast cancer and may represent a potential target for antitumor therapy.展开更多
BACKGROUND Poliomyelitis is an acute infection caused by an enterovirus,which primarily infects the human gastrointestinal tract.In general,patients with polio have no association with the occurrence of cancer.The pre...BACKGROUND Poliomyelitis is an acute infection caused by an enterovirus,which primarily infects the human gastrointestinal tract.In general,patients with polio have no association with the occurrence of cancer.The present case study presents a rare case of poliomyelitis combined with primary breast cancer.CASE SUMMARY A 61-year-old woman who was diagnosed with poliomyelitis at 5 years old and confirmed invasive breast cancer by core needle biopsy(CNB)after hospitalization.The patient received a modified radical mastectomy and four cycles of chemotherapy with the TC(docetaxel and cyclophosphamide)regimen.The patient was also prescribed endocrine therapy without radiotherapy after chemotherapy.The patient had no evidence of lymphedema in the right upper extremities and no evidence of either regression or distant metastasis at the 1-year follow-up.CONCLUSION The pectoral muscles of patients with polio are easily damaged in traumatic procedures,such as CNB,local anesthesia for tumor excision,and general anesthesia for surgery.A CNB,modified radical mastectomy,and four cycles of TC chemotherapy were successfully completed for the present case and the adverse reactions were found to be tolerable.This case may indicate the relationship between breast cancer and polio,and the examination and treatment methods used could be used as a guide for similar cases in the future.展开更多

These results demonstrated that transcranial magnetic stimulation and rehabilitation training altered structural parameters of pyramidal cell dendrites and synapses in the non-injured cerebral hemisphere sensorimotor cortex, thereby improving the ability to compensate for neurological functions in rats following cerebral infarction.
文摘Objective:To explore the application effect of constructing professional teaching staff in low-level training in operating room,so as to further optimize the teaching strength of operating room in our hospital and improve the training effect of junior nurses.Methods:Thirty-eight low-level nurses in the operating room of a third-class hospital in Yantai were selected for half a year's nurse training.With theoretical scores,overall nursing performance,nurses'self-awareness evaluation system and nurses'satisfaction with tutors as evaluation criteria,and based on the selection of high-quality teachers,various evaluation indexes before and after the training of low-level nurses in the operating room were compared and evaluated through the cultivation of practical teaching teachers'ability and the application of a series of teaching methods based on the change of c ompetence-based education(CBE)teaching mode,the application of guided learning interactive canadian education(BOPPPS)teaching model and p roblem-b ased l earning(PBL)teaching method.Results:After the training,the examination scores of low-level nurses were significantly improved(P<0.05),the teaching quality was highly recognized by low-level nurses,the quality of low-level nurses was improved,and patients'satisfaction with nurses was improved.Conclusion:It is of great significance to assist the construction of professional teachers in evidence-based medicine.Through the training of practical teaching teachers'ability and the application of a series of teaching methods based on the change of CBE teaching model,the application of BOPPPS teaching model and PBL teaching method,the training results of nurses have been significantly improved and improved,which is worthy of clinical reference and promotion.

文摘Stem cell transplantation has brought new hope for the treatment of neurological diseases.The key to stem cell therapy lies in inducing the specific differentiation of stem cells into nerve cells.Because the differentiation of stem cells in vitro and in vivo is affected by multiple factors,the final differentiation outcome is strongly associated with the microenvironment in which the stem cells are located.Accordingly,the optimal microenvironment for inducing stem cell differentiation is a hot topic.EGb761 is extracted from the leaves of the Ginkgo biloba tree.It is used worldwide and is becoming one of the focuses of stem cell research.Studies have shown that EGb761 can antagonize oxygen free radicals,stabilize cell membranes,promote neurogenesis and synaptogenesis,increase the level of brain-derived neurotrophic factors,and replicate the environment required during the differentiation of stem cells into nerve cells.This offers the possibility of using EGb761 to induce the differentiation of stem cells,facilitating stem cell transplantation.To provide a comprehensive reference for the future application of EGb761 in stem cell therapy,we reviewed studies investigating the influence of EGb761 on stem cells.These started with the composition and neuropharmacology of EGb761,and eventually led to the finding that EGb761 and some of its important components play important roles in the differentiation of stem cells and the protection of a beneficial microenvironment for stem cell transplantation.

文摘AIM: To characterize the tumor suppressor gene P53 mutations and study the correlation of P53 gene mutation and the expression of P53 protein in cholangiocarcinoma. METHODS: A total of 36 unselected, frozen samples of cholangiocarcinoma were collected, p53 gene status(exon 5-8) and P53 protein were examined by automated sequencing and immunohistochemical staining, combined with the clinical parameters of patients. RESULTS: P53 gene mutations were found in 22 of 36 (61.1%) patients. Nineteen of 36 (52.8%) patients were positive for P53 protein expression. There were significant differences in extent of differentiation and invasion between the positive and negative expression of P53 protein. However, there were no significant differences in pathologic parameters between the mutations and non-mutations. CONCLUSION: The alterations of the P53 gene evaluated by DNA sequence analysis is relatively accurate. Expression of P53 protein could not act as an independent index to estimate the prognosis of cholangiocarcinoma.

文摘Summary: The relationship between tyrosine phosphorylation (TP) and protein expression of insulin receptor (InsR) and insulin resistance (IR) in patients with gestational diabetes mellitus (GDM) was investigated. The InsR expression and TP in skeleton muscle tissue were determined by Western blotting and immunoprecipitation in women with GDM (GDM group, n=22), normal pregnant women (normal pregnancy group, n=22) and normal non-pregnant women (normal non-pregnant group, n=13). Fasting plasma glucose (FPG) and fasting insulin (FINS) were measured by oxidase assay and immunoradioassay. The results showed that the levels of FPG (5.61±0.78 mmol/L), FINS (15.42±5.13 mU/L) and Ho- meostasis model assessment-IR (HOMA-IR) (1.21±0.52) in GDM group were significantly higher than those in normal pregnancy group (4.43±0.46 mmol/L, 10.56±3.07 mU/L and 0.80±0.31 respectively) (P〈0.01). The levels of FINS and HOMA-IR in normal pregnancy group were significantly higher than those in normal non-pregnant group (7.56±2.31 mU/L and 0.47±0.26 respectively) (P〈0.01). There was no significant difference in the InsR expression level among the three groups (P〉0.05). TP of InsR with insulin stimulation was significantly decreased in GDM group (0.20±0.05) as compared with normal pregnancy group (0.26±0.06) (P〈0.01). TP of InsR with insulin stimulation in normal pregnancy group was lower than that in normal non-pregnant group (0.31±0.06) (P〈0.01). TP of InsR with insulin stimu- lation was negatively related with HOMA-IR in GDM group (r=-0.525, P〈0.01). There was no correlation between the protein expression of InsR and HOMA-IR in GDM group (r=-0.236, P〉0.05). It was suggested that there is no significant correlation between the protein expression of InsR in skeletal muscle and IR in GDM, but changes in TP of InsR are associated with IR in GDM.

文摘Objective Iodothyronine deiodinases(DIOs)are important selenoproteins that play a key role in the bone and joint diseases.Osteoarthritis(OA)is the most prevalent joint disease especially in elders.This bioinformatic analysis was performed to explore the role of DIOs in OA pathogenesis.Methods The biological functions of selenoprotein DIOs were analyzed by bioinformatic techniques,mcluding GenCLip 3.0,Database for Annotation,Visualization and Integrated Discovery(DAVID),STRING,Cytoscape,and Network Analyst.The expression of DIOs in the healthy individuals and OA patients was determined by mining OA-related microarray data in the gene expression omnibus(GEO)database of National Center for Biotechnology Information and performing a Meta-analysis of the data with Review Manager 5.3.Results Cluster analysis revealed that the function of the DIOs was associated with thyroid hormone receptor and iodothyronine;GO analysis showed that DIOs were mainly involved in biological processes,such as ethanol metabolism and phenol-containing compound metabolism and primarily involved in the cytochrome P450 metabolism of exogenous organisms and thyroid hormone signaling;SULT1A1 was the core node of the PPI network;miRNAs and thyroid hormones had some iterations with DIO1 and DI02;Meta-analysis showed that DIO3 expression was significantly up-regulated in OA patients(SMD=0.31,95%CI:0.03,0.59,P=0.03).Conclusions The main biological functions of DIOs were closely associated with the regulation of thyroid hormone.And the up-regulated expression of DIO3 may have crucial impact on the occurrence of OA.
文摘Gastrointestinal complications caused by herpes zoster are extremely rare. Here, we described a case of abdominal distention caused by herpes zoster. The patient was a 59-year-old female who suffered from unexplained paroxysmal and a burning pain on the right part of her waist and abdomen, accompanied by abdominal distention. Intestinal pseudo-obstruction was diagnosed by abdominal radiography. Distention of the right abdominal wall was still apparent after one month. In this report, we found that recovery from abdominal distention caused by herpes zoster is difficult and may require surgical intervention.
文摘Endometrial cancer remains to be a major type of malignancy in threatening female life.Molecular insights in advancing our understanding of endometrial tumorigenesis are much needed.We here report that a less-studied protein Dihydropyrimidinase like 3(DPYSL3)is a potent tumor suppressor.DPYSL3 is uniquely regulated by wild type p53(wtp53),and its expression is at the highest level when cells carry wtp53 and are exposed to hypoxia.We reveal that wtp53 can bind DPYSL3 promoter to enhance DPYSL3 expression and in turn,the elevated DPYSL3 can restrain cancer cell proliferation and invasion in vitro and in vivo.Importantly,we observe that DPYSL3 can interfere with MAP kinase pathway,supported by a substantially reduced level of phosphorylated ERK in cells with high expression of DPYSL3.Furthermore,we identify the specific region of DPYSL3 that is responsible for its interaction with MEK and a subsequently reduced activity of ERK.In combination of molecular docking and mutagenesis analysis,we validated that the therapeutic implication of 17 A.A.s of DPYSL3,which can reduce the activity of the MAPK pathway and inhibit endometrial tumor cell growth in vitro and in vivo.Therefore,our study not only demonstrates in-depth understanding of human tumorigenesis,especially endometrial tumor,but also only provides a therapeutic potential to develop an effective tool to fight against human malignancy.
文摘[Objectives] To analyze the dosage,curative effect,and adverse effect of Chidamide in treating peripheral T-cell lymphoma( PTCL). [Methods]A case of treating peripheral T-cell lymphoma by Chidamide was reported,including the treatment process,dosage,curative effect,and adverse effect. Literature review was made and searched in Wanfang Digital Database,China National Knowledge Infrastructure( CNKI),and Pubmed database,using the key word chidamide in Chinese and English separately. Disease and case number of patients,Chidamide observation indicator,curative effect,and adverse effects were recorded in detail. The search was carried out as of September,2016. [Results] It searched 3 articles related to clinical application and 111 cases of patients. [Conclusions] Chidamide has excellent curative effect in treating peripheral T-cell lymphoma and is suitable for clinical application.
文摘[Objectives] To study the expression of Med 19 protein in tongue cancer and its significance. [Methods] Immunohistochemical method was used to detect the expression of Med 19 protein in tongue cancer tissue and paracancerous tissue. [Results]The expression of Med19 protein in tongue cancer tissue was higher than paracancerous tissue and normal tissue. The expression of Med 19 protein was related to the tumor lymph node metastasis. However,there was no significant difference in sex,age,Tumor Node Metastasis( TNM),and differentiation degree. [Conclusions] Med 19 protein participates in incidence and development of tongue cancer and may become diagnostic indicator and therapeutic target for tongue cancer.

文摘BACKGROUND Gastrointestinal stromal tumor(GIST)is a common neoplasm with high rates of recurrence and metastasis,and its therapeutic efficacy is still not ideal.There is an unmet need to find new molecular therapeutic targets for GIST.TATA-boxbinding protein-associated factor 15(TAF15)contributes to the progress of various tumors,while the role and molecular mechanism of TAF15 in GIST progression are still unknown.AIM To explore new molecular therapeutic targets for GIST and understand the biological role and underlying mechanisms of TAF15 in GIST progression.METHODS Proteomic analysis was performed to explore the differentially expressed proteins in GIST.Western blotting and immunohistochemical analysis were used to verify the expression level of TAF15 in GIST tissues and cell lines.Cell counting kit-8,colony formation,wound-healing and transwell assay were executed to detect the ability of TAF15 on cell proliferation,migration and invasion.A xenograft mouse model was applied to explore the role of TAF15 in the progression of GIST.Western blotting was used to detect the phosphorylation level and total level of RAF1,MEK and ERK1/2.RESULTS A total of 1669 proteins were identified as differentially expressed proteins with 762 upregulated and 907 downregulated in GIST.TAF15 was selected for the further study because of its important role in cell proliferation and migration.TAF15 was significantly over expressed in GIST tissues and cell lines.Overexpression of TAF15 was associated with larger tumor size and higher risk stage of GIST.TAF15 knockdown significantly inhibited the cell proliferation and migration of GIST in vitro and suppressed tumor growth in vivo.Moreover,the inhibition of TAF15 expression significantly decreased the phosphorylation level of RAF1,MEK and ERK1/2 in GIST cells and xenograft tissues,while the total RAF1,MEK and ERK1/2 had no significant change.CONCLUSION TAF15 is over expressed in GIST tissues and cell lines.Overexpression of TAF15 was associated with a poor prognosis of GIST patients.TAF15 promotes cell proliferation and migration in GIST via the activation of the RAF1/MEK/ERK signaling pathway.Thus,TAF15 is expected to be a novel latent molecular biomarker or therapeutic target of GIST.
文摘A 46-year-old Chinese woman presented with nausea, recurrent vomiting, and abdominal pain. Gastroduodenal endoscopic examination revealed an oval-shaped submucosal tumor at the prepyloric area on the posterior wall of the stomach. A degenerated gastrointestinal stromal tumor was suspected. Distal gastrectomy was performed and a histological diagnosis of heterotopic pancreas (HPs) was confirmed. The patient had an uneventful postoperative course and was discharged 7 d a^er operation. The patient remains healthy and symptom-free in the follow-up of 6 mo. This is a report of a case of gastric outlet obstruction resulting from pancreatic heterotopia in the gastric antrum in an adult woman.

文摘AIM To evaluate the efficacy of main portal vein stents combined with iodine-125(^(125)Ⅰ) to treat main portal vein tumor thrombus.METHODS From January 1, 2010 to January 1, 2015, 111 patients were diagnosed with liver cancer combined with main portal vein tumor thrombus. They were non-randomly assigned to undergo treatment with transarterial chemoembolization(TACE)/transarterial embolization(TAE) + portal vein stents combined with ^(125)Ⅰ implantation(Group A) and TACE/TAE + portal vein stents only(Group B). After the operation, scheduled follow-up was performed at 6, 12 and 24 mo. The recorded information included clinical manifestations, survival rate, and stent restenosis rate. Kaplan–Meier curves, log-rank test and Cox regression were used for data analyses. RESULTS From January 1, 2010 to January 1, 2015, 54 and 57 patients were allocated to Groups A and B, respectively. The survival rates at 6, 12 and 24 mo were 85.2%, 42.6% and 22.2% in Group A and 50.9%, 10.5% and 0% in Group B. The differences were significant [log rank P < 0.05, hazard ratio(HR): 0.37, 95%CI: 0.24-0.56]. The rates of stent restenosis were 18.5%, 55.6% and 83.3% in Group A and 43.9%, 82.5% and 96.5% in Group B. The differences were significant(log rank P < 0.05, HR: 0.42, 95%CI: 0.27-0.63). Cox regression identified that treatment was the only factor affecting survival rate in this study.CONCLUSION Main portal vein stents combined with ^(125)Ⅰ can significantly improve survival rate and reduce the rate of stent restenosis.

文摘Peripheral nerve injuries occur as the result of sudden trauma and lead to reduced quality of life.The peripheral nervous system has an inherent capability to regenerate axons.However,peripheral nerve regeneration following injury is generally slow and incomplete that results in poor functional outcomes such as muscle atrophy.Although conventional surgical procedures for peripheral nerve injuries present many benefits,there are still several limitations including scarring,difficult accessibility to donor nerve,neuroma formation and a need to sacrifice the autologous nerve.For many years,other therapeutic approaches for peripheral nerve injuries have been explored,the most notable being the replacement of Schwann cells,the glial cells responsible for clearing out debris from the site of injury.Introducing cultured Schwann cells to the injured sites showed great benefits in promoting axonal regeneration and functional recovery.However,there are limited sources of Schwann cells for extraction and difficulties in culturing Schwann cells in vitro.Therefore,novel therapeutic avenues that offer maximum benefits for the treatment of peripheral nerve injuries should be investigated.This review focused on strategies using mesenchymal stem cells to promote peripheral nerve regeneration including exosomes of mesenchymal stem cells,nerve engineering using the nerve guidance conduits containing mesenchymal stem cells,and genetically engineered mesenchymal stem cells.We present the current progress of mesenchymal stem cell treatment of peripheral nerve injuries.

文摘Objective:Although T-cell immunoglobulin and mucin-domain containing molecule-3(Tim-3)has been recognized as a promising target for cancer immunotherapy,its exact role in breast cancer has not been fully elucidated.Methods:Tim-3 gene expression in breast cancer and its prognostic significance were analyzed.Associated mechanisms were then explored in vitro by establishing Tim-3-overexpressing breast cancer cells.Results:In a pooled analysis of The Cancer Genome Atlas(TCGA)database,Tim-3 gene expression levels were significantly higher(P<0.001)in breast cancer tissue,compared with normal tissues.Tim-3 was a prognosis indicator in breast cancer patients[relapse-free survival(RFS),P=0.004;overall survival(OS),P=0.099].Tim-3 overexpression in Tim-3 low breast cancer cells promoted aggressiveness of breast cancer cells,as evidenced by enhanced proliferation,migration,invasion,tight junction deterioration and tumor-associated tubal formation.Tim-3 also enhanced cellular resistance to paclitaxel.Furthermore,Tim-3 exerted its function by activating the NF-κB/STAT3 signalling pathway and by regulating gene expression[cyclin D1(CCND1),C-Myc,matrix metalloproteinase-1(MMP1),TWIST,vascular endothelial growth factor(VEGF)upregulation,concomitant with Ecadherin downregulation).Lastly,Tim-3 downregulated tight junction-associated molecules zona occludens(ZO)-2,ZO-1 and occludin,which may further facilitate tumor progression.Conclusions:Tim-3 plays an oncogenic role in breast cancer and may represent a potential target for antitumor therapy.
文摘BACKGROUND Poliomyelitis is an acute infection caused by an enterovirus,which primarily infects the human gastrointestinal tract.In general,patients with polio have no association with the occurrence of cancer.The present case study presents a rare case of poliomyelitis combined with primary breast cancer.CASE SUMMARY A 61-year-old woman who was diagnosed with poliomyelitis at 5 years old and confirmed invasive breast cancer by core needle biopsy(CNB)after hospitalization.The patient received a modified radical mastectomy and four cycles of chemotherapy with the TC(docetaxel and cyclophosphamide)regimen.The patient was also prescribed endocrine therapy without radiotherapy after chemotherapy.The patient had no evidence of lymphedema in the right upper extremities and no evidence of either regression or distant metastasis at the 1-year follow-up.CONCLUSION The pectoral muscles of patients with polio are easily damaged in traumatic procedures,such as CNB,local anesthesia for tumor excision,and general anesthesia for surgery.A CNB,modified radical mastectomy,and four cycles of TC chemotherapy were successfully completed for the present case and the adverse reactions were found to be tolerable.This case may indicate the relationship between breast cancer and polio,and the examination and treatment methods used could be used as a guide for similar cases in the future.
