Glabridin is the main ingredient of hydrophobic fraction in licorice extract and has been shown to have anti-melanogenesis activity in skins.However,the underlying mechanism(s)remain not completely understood.The aim ...Glabridin is the main ingredient of hydrophobic fraction in licorice extract and has been shown to have anti-melanogenesis activity in skins.However,the underlying mechanism(s)remain not completely understood.The aim of this study is thus to elucidate the possible mechanisms related to the melanogenesis suppression by glabridin in cultured B16 murine melanoma cells and in UVA radiation induced hyperpigmentation model of BALB/c mice as well.Molecular docking simulations revealed that between catalytic core residues and the compound.The treatment by glabridin significantly downregulated both transcriptional and/or protein expression of melanogenesis-related factors including melanocyte stimulating hormone receptor(MC1R),microphthalmia-associated transcription factor(MITF),tyrosinase(TYR),TYR-related protein-1(TRP-1)and TRP-2 in B16 cells.Both PKA/MITF and MAPK/MITF signaling pathways were found to be involved in the suppression of melanogenesis by glabridin in B16 cells.Also in vivo glabridin therapy significantly reduced hyperpigmentation,epidermal thickening,roughness and inflammation induced by frequent UVA exposure in mice skins,thus beneficial for skin healthcare.These data further look insights into the molecular mechanisms of melanogenesis suppression by glabridin,rationalizing the application of the natural compound for skin healthcare.展开更多
Hematopoietic stem cell transplantation has become a curative choice of many hematopoietic malignancy,but graft-vs-host disease(GVHD)has limited the survival quality and overall survival of hematopoietic stem cell tra...Hematopoietic stem cell transplantation has become a curative choice of many hematopoietic malignancy,but graft-vs-host disease(GVHD)has limited the survival quality and overall survival of hematopoietic stem cell transplantation.Understanding of the immune cells’reaction in pathophysiology of GVHD has improved,but a review on the role of macrophages in GVHD is still absent.Studies have observed that macrophage infiltration is associated with GVHD occurrence and development.In this review,we summarize and analyze the role of macrophages in GVHD based on pathophysiology of acute and chronic GVHD,focusing on the macrophage recruitment and infiltration,macrophage polarization,macrophage secretion,and especially interaction of macrophages with other immune cells.We could conclude that macrophage recruitment and infiltration contribute to both acute and chronic GVHD.Based on distinguishing pathology of acute and chronic GVHD,macrophages tend to show a higher M1/M2 ratio in acute GVHD and a lower M1/M2 ratio in chronic GVHD.However,the influence of dominant cytokines in GVHD is controversial and inconsistent with macrophage polarization.In addition,interaction of macrophages with alloreactive T cells plays an important role in acute GVHD.Meanwhile,the interaction among macrophages,B cells,fibroblasts,and CD4+T cells participates in chronic GVHD development.展开更多
BACKGROUND Acetaminophen overdose is the most frequent cause of drug-induced liver failure in developed countries.Substantial progress has been made in understanding the mechanism of hepatocellular injury,but N-acetyl...BACKGROUND Acetaminophen overdose is the most frequent cause of drug-induced liver failure in developed countries.Substantial progress has been made in understanding the mechanism of hepatocellular injury,but N-acetylcysteine remains the only effective treatment despite its short therapeutic window.Thus,other hepatoprotective drugs are needed for the delayed treatment of acetaminopheninduced hepatotoxicity.Our interest focused on glycyrrhizin for its role as an inhibitor of high mobility group box 1(HMGB1)protein,a member of the family of damage-associated molecular pattern,known to play an important pathological role in various diseases.AIM To investigate the efficacy of the N-acetylcysteine/glycyrrhizin combination compared to N-acetylcysteine alone in the prevention of liver toxicity.METHODS Eight-week-old C57BL/6J wild-type female mice were used for all our experiments.Mice fasted for 15 h were treated with acetaminophen(500 mg/kg)or vehicle(phosphate-buffered saline)by intraperitoneal injection and separated into the following groups:Glycyrrhizin(200 mg/kg);N-acetylcysteine(150 mg/kg);and N-acetylcysteine/glycyrrhizin.In all groups,mice were sacrificed 12 h following acetaminophen administration.The assessment of hepatotoxicity was performed by measuring plasma levels of alanine aminotransferase,aspartate aminotransferase and lactate dehydrogenase.Hepatotoxicity was also evaluated by histological examination of hematoxylin and eosin-stained tissues sections.Survival rates were compared between various groups using Kaplan-Meier curves.RESULTS Consistent with data published in the literature,we confirmed that intraperitoneal administration of acetaminophen(500 mg/kg)in mice induced severe liver injury as evidenced by increases in alanine aminotransferase,aspartate aminotransferase and lactate dehydrogenase but also by liver necrosis score.Glycyrrhizin administration was shown to reduce the release of HMGB1 and significantly decreased the severity of liver injury.Thus,the co-administration of glycyrrhizin and N-acetylcysteine was investigated.Administered concomitantly with acetaminophen,the combination significantly reduced the severity of liver injury.Delayed administration of the combination of drugs,2 h or 6 h after acetaminophen,also induced a significant decrease in hepatocyte necrosis compared to mice treated with N-acetylcysteine alone.In addition,administration of N-acetylcysteine/glycyrrhizin combination was associated with an improved survival rate compared to mice treated with only N-acetylcysteine.CONCLUSION We demonstrate that,compared to N-acetylcysteine alone,co-administration of glycyrrhizin decreases the liver necrosis score and improves survival in a murine model of acetaminophen-induced liver injury.Our study opens a potential new therapeutic pathway in the prevention of acetaminophen hepatotoxicity.展开更多

Failure was defned as: (1) non-response [HCV-RNA remained detectable during treatment, at end of treatment (EOT)]; and (2) relapse (HCV-RNA suppressed at EOT but detectable thereafter). Sequencing analysis was performed to describe prevalence of drug class-specifc RAS. Factors associated with failure were determined using logistic regression models.RESULTSAmong 559 patients, 77% had suppressed plasmaHIV-RNA 〈 50 copies/mL at DAA treatment initiation41% were cirrhotic, and 68% were HCV treatmentexperienced. Virological treatment failures occurred in22 patients and were mainly relapses (17, 77%) thenundefined failures (3, 14%) and non-responses (29%). Mean treatment duration was 16 wk overall. Posttreatment NS3, NS5A or NS5B RAS were detected in10/14 patients with samples available for sequencinganalysis. After adjustment for age, sex, ribavirin useHCV genotype and treatment duration, low platelecount was the only factor signifcantly associated with ahigher risk of failure (OR: 6.5; 95%CI: 1.8-22.6). CONCLUSIONOnly 3.9% HIV-HCV coinfected patients failed DAAregimens and RAS were found in 70% of those failingLow platelet count was independently associated withvirological failure.展开更多
The control of light–matter coupling at the single electron level is currently a subject of growing interest for the development of novel quantum devices and for studies and applications of quantum electrodynamics.In...The control of light–matter coupling at the single electron level is currently a subject of growing interest for the development of novel quantum devices and for studies and applications of quantum electrodynamics.In the terahertz(THz)spectral range,this raises the particular and difficult challenge of building electromagnetic resonators that can conciliate low mode volume and high quality factor.Here,we report on hybrid THz cavities based on ultrastrong coupling between a Tamm cavity and an LC circuit metamaterial and show that they can combine high quality factors of up to Q=37 with a deep-subwavelength mode volume of V=3.2×10^(-4)λ^(3).Our theoretical and experimental analysis of the coupled mode properties reveals that,in general,the ultrastrong coupling between a metamaterial and a Fabry–Perot cavity is an effective tool to almost completely suppress radiative losses and,thus,ultimately limit the total losses to the losses in the metallic layer.These Tamm cavity-LC metamaterial coupled resonators open a route toward the development of single photon THz emitters and detectors and to the exploration of ultrastrong THz light–matter coupling with a high degree of coherence in the few to single electron limit.展开更多
Infrared imaging is a crucial technique in a multitude of applications,including night vision,autonomous vehicle navigation,optical tomography,and food quality control.Conventional infrared imaging technologies,howeve...Infrared imaging is a crucial technique in a multitude of applications,including night vision,autonomous vehicle navigation,optical tomography,and food quality control.Conventional infrared imaging technologies,however,require the use of materials such as narrow bandgap semiconductors,which are sensitive to thermal noise and often require cryogenic cooling.We demonstrate a compact all-optical alternative to perform infrared imaging in a metasurface composed of GaAs semiconductor nanoantennas,using a nonlinear wave-mixing process.We experimentally show the upconversion of short-wave infrared wavelengths via the coherent parametric process of sum-frequency generation.In this process,an infrared image of a target is mixed inside the metasurface with a strong pump beam,translating the image from the infrared to the visible in a nanoscale ultrathin imaging device.Our results open up new opportunities for the development of compact infrared imaging devices with applications in infrared vision and life sciences.展开更多
Background:Tauopathies comprise a family of neurodegenerative disorders including Alzheimer’s disease for which there is an urgent and unmet need for disease-modifying treatments.Tauopathies are characterized by path...Background:Tauopathies comprise a family of neurodegenerative disorders including Alzheimer’s disease for which there is an urgent and unmet need for disease-modifying treatments.Tauopathies are characterized by pathological tau hyperphosphorylation,which has been shown to correlate tightly with disease progression and memory loss in patients suffering from Alzheimer’s disease.We recently demonstrated an essential requirement for 3-O-sulfated heparan sulfate in pathological tau hyperphosphorylation in zebrafish,a prominent model organism for human drug discovery.Here,we investigated whether in vivo treatment with surfen or its derivatives oxalyl surfen and hemisurfen,small molecules with heparan sulfate antagonist properties,could mitigate tau hyperphosphorylation and neuronal deficits in a zebrafish model of tauopathies.Results:In vivo treatment of Tg[HuC::hTau^(P301L);DsRed]embryos for 2 days with surfen or oxalyl surfen significantly reduced the accumulation of the pThr181 tau phospho-epitope measured by ELISA by 30%and 51%,respectively.Western blot analysis also showed a significant decrease of pThr181 and pSer396/pSer404 in embryos treated with surfen or oxalyl surfen.Immunohistochemical analysis further confirmed that treatment with surfen or oxalyl surfen significantly decreased the AT8 tau epitope in spinal motoneurons.In addition,in vivo treatment of Tg[HuC::hTau^(P301L);DsRed]embryos with surfen or oxalyl surfen significantly rescued spinal motoneuron axon-branching defects and,as a likely consequence,the impaired stereotypical touch-evoked escape response.Importantly,treatment with hemisurfen,a surfen derivative devoid of heparan sulfate antagonist activity,does not affect tau hyperphosphorylation,nor neuronal or behavioural deficits in Tg[HuC::hTau^(P301L);DsRed]embryos.Conclusion:Our findings demonstrate for the first time that surfen,a well-tolerated molecule in clinical settings,and its derivative,oxalyl surfen,could mitigate or delay neuronal defects in tauopathies,including Alzheimer’s disease.展开更多
Optical modeling coupled to experiments show that a microscope operating in reflection mode allows imaging,through solutions or even a microfluidic cover,various kinds of nanoparticles,NPs,over a(reflecting)sensing su...Optical modeling coupled to experiments show that a microscope operating in reflection mode allows imaging,through solutions or even a microfluidic cover,various kinds of nanoparticles,NPs,over a(reflecting)sensing surface,here a gold(Au)surface.Optical modeling suggests that this configuration enables the interferometric imaging of single NPs which can be characterized individually from local change in the surface reflectivity.The interferometric detection improves the optical limit of detection compared to classical configurations exploiting only the light scattered by the NPs.The method is then tested experimentally,to monitor in situ and in real time,the collision of single Brownian NPs,or optical nanoimpacts,with an Au-sensing surface.First,mimicking a microfluidic biosensor platform,the capture of 300 nm FeOx maghemite NPs from a convective flow by a surface-functionalized Au surface is dynamically monitored.Then,the adsorption or bouncing of individual dielectric(100 nm polystyrene)or metallic(40 and 60 nm silver)NPs is observed directly through the solution.The influence of the electrolyte on the ability of NPs to repetitively bounce or irreversibly adsorb onto the Au surface is evidenced.Exploiting such visualization mode of single-NP optical nanoimpacts is insightful for comprehending single-NP electrochemical studies relying on NP collision on an electrode(electrochemical nanoimpacts).展开更多

t Coelomycetous fungi are an artificial taxonomic group which produce conidia inside a cavity i.e.conidiomata.Coelomycetes comprise about,1000 genera and 7000 species,which can be endophytic,pathogenic or saprobic.Tra...t Coelomycetous fungi are an artificial taxonomic group which produce conidia inside a cavity i.e.conidiomata.Coelomycetes comprise about,1000 genera and 7000 species,which can be endophytic,pathogenic or saprobic.Traditional classification of coelomycetes was previously based on morphology,such as the shape of conidiomata and mode of conidiogenesis,while it was treated as a distinct group i.e.Deuteromycotina.Sequence based taxonomic studies has been used to accommodate asexual fungi in a natural classification system,resolve generic boundaries of polyphyletic genera and species complexes,as well as establish asexual-sexual links.Nevertheless,most of genera lack sequence data,thus,morphology based identification is still important when introducing new genera or species.In this paper we illustrate,describe,and provide taxonomic notes for 235 dematiaceous coelomycetous genera,including five new genera viz.Apiculospora,Didymellocamarosporium,Melanocamarosporium,Melnikia and Paulkirkia.Phylogenetic analyses of combined sequence data are provided to show placements of dematiaceous coelomycetes in Dothideomycetes,Leotiomycetes and Sordariomycetes.One-hundred and fifty-two(65%)of genera have sequence data,thus,their taxonomic placement in a natural classification system,is listed as an outline.However,83 genera still lack sequence data,hence,they are treated as Ascomycota,genera incertae sedis.In addition,separate analyses are provided where better taxonomic resolution is needed.展开更多
Light scattering by a two-dimensional photonic-crystal slab(PCS)can result in marked interference effects associated with Fano resonances.Such devices offer appealing alternatives to distributed Bragg reflectors and f...Light scattering by a two-dimensional photonic-crystal slab(PCS)can result in marked interference effects associated with Fano resonances.Such devices offer appealing alternatives to distributed Bragg reflectors and filters for various applications,such as optical wavelength and polarization filters,reflectors,semiconductor lasers,photodetectors,bio-sensors and non-linear optical components.Suspended PCS also have natural applications in the field of optomechanics,where the mechanical modes of a suspended slab interact via radiation pressure with the optical field of a high-finesse cavity.The reflectivity and transmission properties of a defect-free suspended PCS around normal incidence can be used to couple out-of-plane mechanical modes to an optical field by integrating it in a free-space cavity.Here we demonstrate the successful implementation of a PCS reflector on a high-tensile stress Si_(3)N_(4) nanomembrane.We illustrate the physical process underlying the high reflectivity by measuring the photonic-crystal band diagram.Moreover,we introduce a clear theoretical description of the membrane scattering properties in the presence of optical losses.By embedding the PCS inside a high-finesse cavity,we fully characterize its optical properties.The spectrally,angular-and polarization-resolved measurements demonstrate the wide tunability of the membrane’s reflectivity,from nearly 0 to 99.9470±0.0025%,and show that material absorption is not the main source of optical loss.Moreover,the cavity storage time demonstrated in this work exceeds the mechanical period of low-order mechanical drum modes.This so-called resolved-sideband condition is a prerequisite to achieve quantum control of the mechanical resonator with light.展开更多
Macroscale fluorescence imaging is increasingly used to observe biological samples.However,it may suffer from spectral interferences that originate from ambient light or autofluorescence of the sample or its support.I...Macroscale fluorescence imaging is increasingly used to observe biological samples.However,it may suffer from spectral interferences that originate from ambient light or autofluorescence of the sample or its support.In this manuscript,we built a simple and inexpensive fluorescence macroscope,which has been used to evaluate the performance of Speed OPIOM(Out of Phase Imaging after Optical Modulation),which is a reference-free dynamic contrast protocol,to selectively image reversibly photoswitchable fluorophores as labels against detrimental autofluorescence and ambient light.By tuning the intensity and radial frequency of the modulated illumination to the Speed OPIOM resonance and adopting a phase-sensitive detection scheme that ensures noise rejection,we enhanced the sensitivity and the signal-to-noise ratio for fluorescence detection in blot assays by factors of 50 and 10,respectively,over direct fluorescence observation under constant illumination.Then,we overcame the strong autofluorescence of growth media that are currently used in microbiology and realized multiplexed fluorescence observation of colonies of spectrally similar fluorescent bacteria with a unique configuration of excitation and emission wavelengths.Finally,we easily discriminated fluorescent labels from the autofluorescent and reflective background in labeled leaves,even under the interference of incident light at intensities that are comparable to sunlight.The proposed approach is expected to find multiple applications,from biological assays to outdoor observations,in fluorescence macroimaging.展开更多

Aim:Angiogenesis is observed in acute myeloid leukemia(AML).AML cells abnormally proliferate and are resistant to death.Positive regulators of angiogenesis,VEGF-A and matrix metalloproteinases(MMPs)2 and 9 are markers...Aim:Angiogenesis is observed in acute myeloid leukemia(AML).AML cells abnormally proliferate and are resistant to death.Positive regulators of angiogenesis,VEGF-A and matrix metalloproteinases(MMPs)2 and 9 are markers of disease status in AML.The natural phloroglucinol hyperforin(HF)displays antitumoral properties of potential pharmacological interest.Herein,we investigated the effects of HF on MMP-2/9 and VEGF-A expression and survival of primary AML cells.Methods:Blood and bone marrow samples were collected in 45 patients with distinct subtypes defined by French American British classification,i.e.,M0,M1,M2,M3,M4,and M5.Levels of MMPs and VEGF-A in leukemic blood cells and culture supernatants were determined by RT-PCR,ELISA,and gelatin zymography(MMPs).The balance between cell death and survival was assessed by flow cytometry with analysis of phosphatidylserine externalization and caspase-3 activation.Results:The administration of HF promoted a caspase-associated apoptosis in primary AML blasts(from blood and bone marrow),but not normal blood cells and monocytes.In addition,HF inhibited the levels of secreted proMMP-2,proMMP-9,and VEGF-A without altering transcripts.The induction of apoptosis by HF significantly paralleled the inhibition of MMP-2/9 and VEGF-A release by HF.No differences were seen in response to the deleterious effects of HF between AML cells of distinct subtypes.Conclusion:Our results suggest that HF,through its proapoptotic and potential antiangiogenic properties(by inhibiting MMP-2/9 and VEGF-A)on primary AML cells,might be a useful experimental agent,in combination with existing drugs,for new therapeutic approaches in the treatment of this incurable disease.展开更多

文摘Glabridin is the main ingredient of hydrophobic fraction in licorice extract and has been shown to have anti-melanogenesis activity in skins.However,the underlying mechanism(s)remain not completely understood.The aim of this study is thus to elucidate the possible mechanisms related to the melanogenesis suppression by glabridin in cultured B16 murine melanoma cells and in UVA radiation induced hyperpigmentation model of BALB/c mice as well.Molecular docking simulations revealed that between catalytic core residues and the compound.The treatment by glabridin significantly downregulated both transcriptional and/or protein expression of melanogenesis-related factors including melanocyte stimulating hormone receptor(MC1R),microphthalmia-associated transcription factor(MITF),tyrosinase(TYR),TYR-related protein-1(TRP-1)and TRP-2 in B16 cells.Both PKA/MITF and MAPK/MITF signaling pathways were found to be involved in the suppression of melanogenesis by glabridin in B16 cells.Also in vivo glabridin therapy significantly reduced hyperpigmentation,epidermal thickening,roughness and inflammation induced by frequent UVA exposure in mice skins,thus beneficial for skin healthcare.These data further look insights into the molecular mechanisms of melanogenesis suppression by glabridin,rationalizing the application of the natural compound for skin healthcare.

文摘Hematopoietic stem cell transplantation has become a curative choice of many hematopoietic malignancy,but graft-vs-host disease(GVHD)has limited the survival quality and overall survival of hematopoietic stem cell transplantation.Understanding of the immune cells’reaction in pathophysiology of GVHD has improved,but a review on the role of macrophages in GVHD is still absent.Studies have observed that macrophage infiltration is associated with GVHD occurrence and development.In this review,we summarize and analyze the role of macrophages in GVHD based on pathophysiology of acute and chronic GVHD,focusing on the macrophage recruitment and infiltration,macrophage polarization,macrophage secretion,and especially interaction of macrophages with other immune cells.We could conclude that macrophage recruitment and infiltration contribute to both acute and chronic GVHD.Based on distinguishing pathology of acute and chronic GVHD,macrophages tend to show a higher M1/M2 ratio in acute GVHD and a lower M1/M2 ratio in chronic GVHD.However,the influence of dominant cytokines in GVHD is controversial and inconsistent with macrophage polarization.In addition,interaction of macrophages with alloreactive T cells plays an important role in acute GVHD.Meanwhile,the interaction among macrophages,B cells,fibroblasts,and CD4+T cells participates in chronic GVHD development.

文摘BACKGROUND Acetaminophen overdose is the most frequent cause of drug-induced liver failure in developed countries.Substantial progress has been made in understanding the mechanism of hepatocellular injury,but N-acetylcysteine remains the only effective treatment despite its short therapeutic window.Thus,other hepatoprotective drugs are needed for the delayed treatment of acetaminopheninduced hepatotoxicity.Our interest focused on glycyrrhizin for its role as an inhibitor of high mobility group box 1(HMGB1)protein,a member of the family of damage-associated molecular pattern,known to play an important pathological role in various diseases.AIM To investigate the efficacy of the N-acetylcysteine/glycyrrhizin combination compared to N-acetylcysteine alone in the prevention of liver toxicity.METHODS Eight-week-old C57BL/6J wild-type female mice were used for all our experiments.Mice fasted for 15 h were treated with acetaminophen(500 mg/kg)or vehicle(phosphate-buffered saline)by intraperitoneal injection and separated into the following groups:Glycyrrhizin(200 mg/kg);N-acetylcysteine(150 mg/kg);and N-acetylcysteine/glycyrrhizin.In all groups,mice were sacrificed 12 h following acetaminophen administration.The assessment of hepatotoxicity was performed by measuring plasma levels of alanine aminotransferase,aspartate aminotransferase and lactate dehydrogenase.Hepatotoxicity was also evaluated by histological examination of hematoxylin and eosin-stained tissues sections.Survival rates were compared between various groups using Kaplan-Meier curves.RESULTS Consistent with data published in the literature,we confirmed that intraperitoneal administration of acetaminophen(500 mg/kg)in mice induced severe liver injury as evidenced by increases in alanine aminotransferase,aspartate aminotransferase and lactate dehydrogenase but also by liver necrosis score.Glycyrrhizin administration was shown to reduce the release of HMGB1 and significantly decreased the severity of liver injury.Thus,the co-administration of glycyrrhizin and N-acetylcysteine was investigated.Administered concomitantly with acetaminophen,the combination significantly reduced the severity of liver injury.Delayed administration of the combination of drugs,2 h or 6 h after acetaminophen,also induced a significant decrease in hepatocyte necrosis compared to mice treated with N-acetylcysteine alone.In addition,administration of N-acetylcysteine/glycyrrhizin combination was associated with an improved survival rate compared to mice treated with only N-acetylcysteine.CONCLUSION We demonstrate that,compared to N-acetylcysteine alone,co-administration of glycyrrhizin decreases the liver necrosis score and improves survival in a murine model of acetaminophen-induced liver injury.Our study opens a potential new therapeutic pathway in the prevention of acetaminophen hepatotoxicity.

文摘Most studies focus on the adaptive immune cells in the GVHD pathogenesis,while little is known about innate immune cells in GVHD occurrence and development,especially macrophages.Meanwhile,a higher incidence of graft versus host disease(GVHD)is also found in the elderly patients.Though advances have been made in the modification of macrophages influenced by the inflamm-ageing,there is still no review on the role of macrophages in GVHD and the association between GVHD and the altered macrophages by inflamm-ageing.In this review,we focus on the potential age-related modifications of macrophage in GVHD,which contributes to the change of morbidity and mortality of GVHD.Via literature review,we found that the infiltration of macrophages is associated with GVHD and macrophages are modified in inflamm-ageing state,including the proliferation,migration,phagocytosis,antigen presentation,interaction with other immune cells,and pro-fibrosis.We suppose that altered macrophage functions in inflamm-ageing state contribute to GVHD in elderly patients.
文摘Hepcidin is the hyposideremic hormone regulating iron metabolism.It is a defensin-like disulfide-bonded peptide with antimicrobial activity.The main site of hepcidin production is the liver where its synthesis is modulated by iron,inflammation and erythropoietic signaling.However,hepcidin locally produced in several peripheral organs seems to be an important actor for the maintenance of iron homeostasis in these organs.This review highlights the presence of peripheral hepcidin and its potential functions.Understanding the role of extrahepatic hepcidin could be of great physiological and therapeutic importance for several specific pathologies.

文摘AIMTo describe factors associated with treatment failure and frequency of resistance-associated substitutions (RAS).METHODSHuman immunodefciency virus (HIV)/hepatitis C virus (HCV) coinfected patients starting a first direct-acting antiviral (DAA) regimen before February 2016 and included in the French ANRS CO13 HEPAVIH cohort were eligible. Failure was defned as: (1) non-response [HCV-RNA remained detectable during treatment, at end of treatment (EOT)]; and (2) relapse (HCV-RNA suppressed at EOT but detectable thereafter). Sequencing analysis was performed to describe prevalence of drug class-specifc RAS. Factors associated with failure were determined using logistic regression models.RESULTSAmong 559 patients, 77% had suppressed plasmaHIV-RNA 〈 50 copies/mL at DAA treatment initiation41% were cirrhotic, and 68% were HCV treatmentexperienced. Virological treatment failures occurred in22 patients and were mainly relapses (17, 77%) thenundefined failures (3, 14%) and non-responses (29%). Mean treatment duration was 16 wk overall. Posttreatment NS3, NS5A or NS5B RAS were detected in10/14 patients with samples available for sequencinganalysis. After adjustment for age, sex, ribavirin useHCV genotype and treatment duration, low platelecount was the only factor signifcantly associated with ahigher risk of failure (OR: 6.5; 95%CI: 1.8-22.6). CONCLUSIONOnly 3.9% HIV-HCV coinfected patients failed DAAregimens and RAS were found in 70% of those failingLow platelet count was independently associated withvirological failure.

文摘The control of light–matter coupling at the single electron level is currently a subject of growing interest for the development of novel quantum devices and for studies and applications of quantum electrodynamics.In the terahertz(THz)spectral range,this raises the particular and difficult challenge of building electromagnetic resonators that can conciliate low mode volume and high quality factor.Here,we report on hybrid THz cavities based on ultrastrong coupling between a Tamm cavity and an LC circuit metamaterial and show that they can combine high quality factors of up to Q=37 with a deep-subwavelength mode volume of V=3.2×10^(-4)λ^(3).Our theoretical and experimental analysis of the coupled mode properties reveals that,in general,the ultrastrong coupling between a metamaterial and a Fabry–Perot cavity is an effective tool to almost completely suppress radiative losses and,thus,ultimately limit the total losses to the losses in the metallic layer.These Tamm cavity-LC metamaterial coupled resonators open a route toward the development of single photon THz emitters and detectors and to the exploration of ultrastrong THz light–matter coupling with a high degree of coherence in the few to single electron limit.

文摘Infrared imaging is a crucial technique in a multitude of applications,including night vision,autonomous vehicle navigation,optical tomography,and food quality control.Conventional infrared imaging technologies,however,require the use of materials such as narrow bandgap semiconductors,which are sensitive to thermal noise and often require cryogenic cooling.We demonstrate a compact all-optical alternative to perform infrared imaging in a metasurface composed of GaAs semiconductor nanoantennas,using a nonlinear wave-mixing process.We experimentally show the upconversion of short-wave infrared wavelengths via the coherent parametric process of sum-frequency generation.In this process,an infrared image of a target is mixed inside the metasurface with a strong pump beam,translating the image from the infrared to the visible in a nanoscale ultrathin imaging device.Our results open up new opportunities for the development of compact infrared imaging devices with applications in infrared vision and life sciences.

文摘Background:Tauopathies comprise a family of neurodegenerative disorders including Alzheimer’s disease for which there is an urgent and unmet need for disease-modifying treatments.Tauopathies are characterized by pathological tau hyperphosphorylation,which has been shown to correlate tightly with disease progression and memory loss in patients suffering from Alzheimer’s disease.We recently demonstrated an essential requirement for 3-O-sulfated heparan sulfate in pathological tau hyperphosphorylation in zebrafish,a prominent model organism for human drug discovery.Here,we investigated whether in vivo treatment with surfen or its derivatives oxalyl surfen and hemisurfen,small molecules with heparan sulfate antagonist properties,could mitigate tau hyperphosphorylation and neuronal deficits in a zebrafish model of tauopathies.Results:In vivo treatment of Tg[HuC::hTau^(P301L);DsRed]embryos for 2 days with surfen or oxalyl surfen significantly reduced the accumulation of the pThr181 tau phospho-epitope measured by ELISA by 30%and 51%,respectively.Western blot analysis also showed a significant decrease of pThr181 and pSer396/pSer404 in embryos treated with surfen or oxalyl surfen.Immunohistochemical analysis further confirmed that treatment with surfen or oxalyl surfen significantly decreased the AT8 tau epitope in spinal motoneurons.In addition,in vivo treatment of Tg[HuC::hTau^(P301L);DsRed]embryos with surfen or oxalyl surfen significantly rescued spinal motoneuron axon-branching defects and,as a likely consequence,the impaired stereotypical touch-evoked escape response.Importantly,treatment with hemisurfen,a surfen derivative devoid of heparan sulfate antagonist activity,does not affect tau hyperphosphorylation,nor neuronal or behavioural deficits in Tg[HuC::hTau^(P301L);DsRed]embryos.Conclusion:Our findings demonstrate for the first time that surfen,a well-tolerated molecule in clinical settings,and its derivative,oxalyl surfen,could mitigate or delay neuronal defects in tauopathies,including Alzheimer’s disease.

文摘Optical modeling coupled to experiments show that a microscope operating in reflection mode allows imaging,through solutions or even a microfluidic cover,various kinds of nanoparticles,NPs,over a(reflecting)sensing surface,here a gold(Au)surface.Optical modeling suggests that this configuration enables the interferometric imaging of single NPs which can be characterized individually from local change in the surface reflectivity.The interferometric detection improves the optical limit of detection compared to classical configurations exploiting only the light scattered by the NPs.The method is then tested experimentally,to monitor in situ and in real time,the collision of single Brownian NPs,or optical nanoimpacts,with an Au-sensing surface.First,mimicking a microfluidic biosensor platform,the capture of 300 nm FeOx maghemite NPs from a convective flow by a surface-functionalized Au surface is dynamically monitored.Then,the adsorption or bouncing of individual dielectric(100 nm polystyrene)or metallic(40 and 60 nm silver)NPs is observed directly through the solution.The influence of the electrolyte on the ability of NPs to repetitively bounce or irreversibly adsorb onto the Au surface is evidenced.Exploiting such visualization mode of single-NP optical nanoimpacts is insightful for comprehending single-NP electrochemical studies relying on NP collision on an electrode(electrochemical nanoimpacts).

文摘Particle-bound mercury(PBM)records the oxidation of elemental mercury,of which the main oxidation pathways(Br∙/Cl∙/OH∙/O3)remain unclear,especially in the Southern Hemisphere.Here,we present latitudinal covariations of Hg and Sisotopic anomalies in cross-hemispheric marine aerosols that evidence an equator-to-poleward transition of Hg oxidants from OH∙/O3 in tropics to Br∙/Cl∙in polar regions highlighting thus the presence of distinct oxidation processes producing PBM.The correlations between Hg,S and O-isotopic compositions measured in PBM,sulfates and nitrates respectively within the aerosols highlight the implication of common oxidants in their formations at different latitudes.Our results open a new window to better quantify the present-day atmospheric Hg,S and N budgets and to evaluate the influences of aerosols on climate and ecosystems once the isotopic fractionations associated with each process have been determined.

文摘t Coelomycetous fungi are an artificial taxonomic group which produce conidia inside a cavity i.e.conidiomata.Coelomycetes comprise about,1000 genera and 7000 species,which can be endophytic,pathogenic or saprobic.Traditional classification of coelomycetes was previously based on morphology,such as the shape of conidiomata and mode of conidiogenesis,while it was treated as a distinct group i.e.Deuteromycotina.Sequence based taxonomic studies has been used to accommodate asexual fungi in a natural classification system,resolve generic boundaries of polyphyletic genera and species complexes,as well as establish asexual-sexual links.Nevertheless,most of genera lack sequence data,thus,morphology based identification is still important when introducing new genera or species.In this paper we illustrate,describe,and provide taxonomic notes for 235 dematiaceous coelomycetous genera,including five new genera viz.Apiculospora,Didymellocamarosporium,Melanocamarosporium,Melnikia and Paulkirkia.Phylogenetic analyses of combined sequence data are provided to show placements of dematiaceous coelomycetes in Dothideomycetes,Leotiomycetes and Sordariomycetes.One-hundred and fifty-two(65%)of genera have sequence data,thus,their taxonomic placement in a natural classification system,is listed as an outline.However,83 genera still lack sequence data,hence,they are treated as Ascomycota,genera incertae sedis.In addition,separate analyses are provided where better taxonomic resolution is needed.

文摘Light scattering by a two-dimensional photonic-crystal slab(PCS)can result in marked interference effects associated with Fano resonances.Such devices offer appealing alternatives to distributed Bragg reflectors and filters for various applications,such as optical wavelength and polarization filters,reflectors,semiconductor lasers,photodetectors,bio-sensors and non-linear optical components.Suspended PCS also have natural applications in the field of optomechanics,where the mechanical modes of a suspended slab interact via radiation pressure with the optical field of a high-finesse cavity.The reflectivity and transmission properties of a defect-free suspended PCS around normal incidence can be used to couple out-of-plane mechanical modes to an optical field by integrating it in a free-space cavity.Here we demonstrate the successful implementation of a PCS reflector on a high-tensile stress Si_(3)N_(4) nanomembrane.We illustrate the physical process underlying the high reflectivity by measuring the photonic-crystal band diagram.Moreover,we introduce a clear theoretical description of the membrane scattering properties in the presence of optical losses.By embedding the PCS inside a high-finesse cavity,we fully characterize its optical properties.The spectrally,angular-and polarization-resolved measurements demonstrate the wide tunability of the membrane’s reflectivity,from nearly 0 to 99.9470±0.0025%,and show that material absorption is not the main source of optical loss.Moreover,the cavity storage time demonstrated in this work exceeds the mechanical period of low-order mechanical drum modes.This so-called resolved-sideband condition is a prerequisite to achieve quantum control of the mechanical resonator with light.

文摘Macroscale fluorescence imaging is increasingly used to observe biological samples.However,it may suffer from spectral interferences that originate from ambient light or autofluorescence of the sample or its support.In this manuscript,we built a simple and inexpensive fluorescence macroscope,which has been used to evaluate the performance of Speed OPIOM(Out of Phase Imaging after Optical Modulation),which is a reference-free dynamic contrast protocol,to selectively image reversibly photoswitchable fluorophores as labels against detrimental autofluorescence and ambient light.By tuning the intensity and radial frequency of the modulated illumination to the Speed OPIOM resonance and adopting a phase-sensitive detection scheme that ensures noise rejection,we enhanced the sensitivity and the signal-to-noise ratio for fluorescence detection in blot assays by factors of 50 and 10,respectively,over direct fluorescence observation under constant illumination.Then,we overcame the strong autofluorescence of growth media that are currently used in microbiology and realized multiplexed fluorescence observation of colonies of spectrally similar fluorescent bacteria with a unique configuration of excitation and emission wavelengths.Finally,we easily discriminated fluorescent labels from the autofluorescent and reflective background in labeled leaves,even under the interference of incident light at intensities that are comparable to sunlight.The proposed approach is expected to find multiple applications,from biological assays to outdoor observations,in fluorescence macroimaging.

文摘In this paper we describe the potential of the enhanced X-ray Timing and Polarimetry(eXTP) mission for studies related to accretion flows in the strong field gravity regime around both stellar-mass and supermassive black-holes. eXTP has the unique capability of using advanced "spectral-timing-polarimetry" techniques to analyze the rapid variations with three orthogonal diagnostics of the flow and its geometry, yielding unprecedented insight into the inner accreting regions, the effects of strong field gravity on the material within them and the powerful outflows which are driven by the accretion process.

文摘Aim:Angiogenesis is observed in acute myeloid leukemia(AML).AML cells abnormally proliferate and are resistant to death.Positive regulators of angiogenesis,VEGF-A and matrix metalloproteinases(MMPs)2 and 9 are markers of disease status in AML.The natural phloroglucinol hyperforin(HF)displays antitumoral properties of potential pharmacological interest.Herein,we investigated the effects of HF on MMP-2/9 and VEGF-A expression and survival of primary AML cells.Methods:Blood and bone marrow samples were collected in 45 patients with distinct subtypes defined by French American British classification,i.e.,M0,M1,M2,M3,M4,and M5.Levels of MMPs and VEGF-A in leukemic blood cells and culture supernatants were determined by RT-PCR,ELISA,and gelatin zymography(MMPs).The balance between cell death and survival was assessed by flow cytometry with analysis of phosphatidylserine externalization and caspase-3 activation.Results:The administration of HF promoted a caspase-associated apoptosis in primary AML blasts(from blood and bone marrow),but not normal blood cells and monocytes.In addition,HF inhibited the levels of secreted proMMP-2,proMMP-9,and VEGF-A without altering transcripts.The induction of apoptosis by HF significantly paralleled the inhibition of MMP-2/9 and VEGF-A release by HF.No differences were seen in response to the deleterious effects of HF between AML cells of distinct subtypes.Conclusion:Our results suggest that HF,through its proapoptotic and potential antiangiogenic properties(by inhibiting MMP-2/9 and VEGF-A)on primary AML cells,might be a useful experimental agent,in combination with existing drugs,for new therapeutic approaches in the treatment of this incurable disease.
