Therefore, maintaining sternness while expanding th...Despite their potential applications in future regenerative medicine, periodontal ligament stem cells(PDLSCs) are difficult to obtain in large amounts from patients. Therefore, maintaining sternness while expanding the cell numbers for medical use is the key to transitioning PDLSCs from the bench to the clinic. Lysophosphatidic acid(LPA), which is present in the human body and saliva, is a signaling molecule derived from phospholipids. In this study, we examined the effects of LPA on sternness maintenance in human PDLSCs. Several spindle-shaped and fibroblast-like periodontal ligament stem-like cell lines were established from PDLSC isolation. Among these cell lines, the most morphologically appropriate cell line was characterized. The expression levels of OCT4, NANOG(a stem cell marker), and CD90(a mesenchymal stem cell marker) were high. However, CD73(a negative marker of mesenchymal stem cells) expression was not observed. Notably, immunofluorescence analysis identified the expression of STRO-1, CD146(a mesenchymal stem cell marker), and sex determining region Y-box 2 at the protein level. In addition, lipid droplets were stained by Oil red O after the induction of adipogenesis for 21 days, and mineralized nodules were stained by Alizarin Red S after the induction of osteogenesis for 14 days. Alkaline phosphate staining also demonstrated the occurrence of osteogenesis. In summary, we established a human PDLSC line, which could be applied as a cell source for tissue regeneration in dental patients. However, further studies are needed to determine the detailed effects of LPA on PDLSCs.展开更多
Bisphenol-A(BPA) has been considered as an endocrine disrupting chemical(EDC) because it can exert estrogenic properties.For bisphenol-S(BPS) and bisphenol-F(BPF) that are BPA analogs and substitutes,their ris...Bisphenol-A(BPA) has been considered as an endocrine disrupting chemical(EDC) because it can exert estrogenic properties.For bisphenol-S(BPS) and bisphenol-F(BPF) that are BPA analogs and substitutes,their risk to estrogendependent cancer has been reported rarely compared with the numerous cases of BPA.In this study,we examined whether BPA,BPS,and BPF can lead to the proliferation,migration,and epithelial mesenchymal transition(EMT) of MCF-7 clonal variant(MCF-7 CV) breast cancer cells expressing estrogen receptors(ERs).In a cell viability assay,BPA,BPS,and BPF significantly increased proliferation of MCF-7 CV cells compared to control(DMSO) as did17β-estradiol(E2).In Western blotting assay,BPA,BPS,and BPF enhanced the protein expression of cell cycle progression genes such as cyclin D1 and E1.In addition,MCF-7 CV cells lost cell to cell contacts and acquired fibroblast-like morphology by the treatment of BPA,BPS,or BPF for 24 hours.In cell migration assay,BPA,BPS,and BPF accelerated the migration capability of MCF-7 CV cells as did E2.In relation with the EMT process,BPA,BPS,and BPF increased the protein expression of N-cadherin,while they decreased the protein expression of Ecadherin.When BPA,BPS,and BPF were co-treated with ICI 182,780,an ER antagonist,proliferation effects were reversed,the expression of cyclin D1 and cyclin E1 was downregulated,and the altered cell migration and expression of N-cadherin and E-cadherin by BPA,BPS,and BPF were restored to the control level.Thus,these results imply that BPS and BPF also have the risk of breast cancer progression as much as BPA in the induction of proliferation and migration of MCF-7 CV cells by regulating the protein expression of cell cycle-related genes and EMT markers via the ER-dependent pathway.展开更多

The aim of the present study was to investigate the effects of zeaxanthin supplementation on in vitro maturation of porcine embryo development. We investigated nuclear maturation, intracellular glutathione (GSH), and reactive oxygen species (ROS) levels during in vitro maturation, and subsequent embryonic development following parthenogenetic activation and in vitro fertilization OVF). The oocytes were maturated and used at the metaphase II stage. After 42 hours of in vitro maturation, the zeaxanthin-treated group (0.5 μmol/L) showed significant increases in nuclear maturation (89.6%) than the control group (83.4%) (P 〈 0.05). The intracellular GSH levels increased significantly (P 〈 0.05) as zeaxanthin concentrations increased; ROS generation levels decreased with increased zeaxanthin concentrations, but there were no significant differences. There were no significant differences in subsequent embryonic development, cleavage rate, blastocyst stage rate, and total blastocyst cell numbers following parthenogenetic activation and IVF when in vitro maturation media was supplemented with zeaxanthin. These results suggest that treatment with zeaxanthin during in vitro maturation improved the nuclear maturation of porcine oocytes by increasing the intracellular GSH level, thereby slightly decreasing the intracellular ROS level.展开更多

Results: The results showed that neem oil extract significantly reduced the viability in all selected cancer cells treated with varying concentrations of extract as compared with untreated cells and had less effect on noncancerous cell lines. It significantly increased the percentage of necrotic and apoptotic cells, and caspases 3, 8 and 9 activities in all cancer cells treated with extract as compared with untreated cells whereas no effect on noncancerous cell lines. It suggested that neem oil extract exerted a higher cytotoxic effect on cancer cells than normal cells and lower concentration induced apoptosis only in cancer cells.One of the apoptosis-inducing mechanism was through the activation of caspases signaling pathways. Conclusion: Conclusively, it implies that neem oil extract may contain one or more potential agents that can be used as a safe and effective anticancer therapy.展开更多

Sprague- Dawley rats were fed a diet containing the oils or trans-fat, and then body weights, blood lipids, and effects on brain infarction and physical dysfunction induced by middle cerebral artery occlusion (MCAO) were analyzed. All the oils and trans-fat, except perilla oil, significantly increased body fats and body weight gain. Sesame oil and trans-fat specifically increased blood cholesterols and triglycerides, respectively, while perilla oil decreased both cholesterols and triglycerides. Perilla oil not only attenuated cerebral infarction, but also restored locomotor activity and rota-rod performances of MCAO rats. It is suggested that perilla oil among oils and fats could be the first choice to reduce the risk of metabolic syndrome and ischemic stroke.展开更多

We investigated the capacities of a hydrogen-enriched water(HW) eliminating carbon nanoparticles(CNP) and carbon microparticles(CMP) from the lungs and blood, respectively. In CNP-elimination test, rats were orally administered with purified water(PW) or HW(10 or 30 mL/kg/day) for 10 weeks. At the time point of 4 weeks, the rats were challenged with intratracheal instillation of CNP(4 mg). CNP accumulated in the airways and alveoli, and induced inflammatory lesions. Such pneumoconiosis was markedly improved by feeding HW, while PW was ineffective. CNP-induced pneumoconiosis caused systemic hematological alterations, decreasing major inflammatory cells, but markedly increasing eosinophils,indicative of an allergic reaction, which were attenuated by treatment with HW. Such PM-eliminating and antiallergic effects of HW reduced body burden as confirmed from the facilitated recovery of body and lung weights. In CMP-clearance test, mice were orally administered with PW or HW for 7 days, and intravenously injected with CMP(300 mg/kg). CMP was rapidly eliminated from the blood in HW-fed mice. Indeed, the phagocytic indices increased to 3.5 and 6.7 folds at 10 and 30 mL/kg of HW, in comparison with a negligible effect of PW. As a mechanism study,only HW significantly inhibited lipid peroxidation in vitro Fenton reaction-mediated ·OH-generating system.Collectively, the results indicate that HW not only effectively eliminated PM from the lungs and blood by enhancing phagocytic activity, but also attenuated the lung injuries by inhibiting lipid peroxidation.展开更多

To evaluate the...Natural volatile organic compounds(VOCs) extracted from conifers such as P. koraiensis and L. kaempferi have long been studied for their anti-oxidant, anti-proliferative, and anti-inflammatory effects. To evaluate the anti-inflammatory effects of VOCs from P. koraiensis and L. kaempferi, lipopolysaccharide(LPS) was administered to generate a mouse model for inflammation by the nasal route to the lungs and intraperitoneally to the whole body. VOCs of P. koraiensis and L. kaempferi were exposed to the mice by standardized wood panels with closed system. Increased levels of serum IgE and PGE2 were observed after exposure to dexamethasone and VOCs. We further determined the expression levels of inflammatory cytokine mRNA in the LPS-induced inflammation model by the reverse transcription quantitative polymerase chain reaction. Furthermore, the levels of cyclooxygenase-2, tumor necrosis factor-α, interleukin-1β, and interleukin-13 were determined in peripheral blood mononuclear cells. Those inflammatory cytokines and the key enzyme for inflammation cyclooxygenase-2 expression in PBMCs were strongly reversed by dexamethasone and VOCs. Lung tissues after nasal LPS exposure showed increased cytokine mRNA expressions which were suppressed by treatment with dexamethasone and VOCs. Furthermore, the damage induced by LPS was attenuated by dexamethasone and VOCs. In conclusion, the results from the present study indicate that VOCs of P. koraiensis and L. kaempferi have a therapeutic potential in the treatment or prevention of local and systemic inflammation due to their immunosuppressive effects.展开更多

Pregnant rats were fed the soybean milk(5% or 100% in drinking water)from gestational day(GD) 6 to parturition or to post-natal day(PND) 56. Specifically, the rats were divided into 4 groups: the control group(drinking water), the GD5% group(5% soybean milk during only the GD period), the GDPND5% group(5% soybean milk during the GD and PND periods), and the GD-PND100% group(100% soybean milk instead of water during the GD and PND periods). During the gestational, lactational, and developmental periods, the reproductive and developmental parameters of dams and offspring were observed. Feeding soybean milk did not affect the birth and physical development of both male and female offspring. At PND57, the weights of the testes and epididymides of F1 males significantly increased by feeding a high concentration of the soybean milk(GDPND100%). In addition, feeding of the soybean milk during both the GD and PND periods(GD-PND5% and GDPND100%) enhanced the sperm counts and motility. The results indicate that soybean milk is safe for embryos,fetuses, and offspring, and improves the post-generational development of male reproductive function.展开更多
Purpose: To determine if cats with chronic kidney disease (CKD) would willingly consume an oral nutritional supplement formula (NS-CKD) and to assess associated effects on select clinical and biochemical parameters. M...Purpose: To determine if cats with chronic kidney disease (CKD) would willingly consume an oral nutritional supplement formula (NS-CKD) and to assess associated effects on select clinical and biochemical parameters. Methods: Client-owned cats with CKD classified as International Renal Interest Society (IRIS) stage 2 (n = 7), IRIS stage 3 (n = 12), or IRIS stage 4 (1 cat) were classified by the owners as having normal or variable appetites. The cats were offered 30 ml NS-CKD for 14 days concurrently with a meal in a separate bowl and the amount of the NS-CKD consumed daily was recorded. Bodyweight, physical examination, and serum biochemical profiles were assessed on Days 0, 7, and 14. Results: Greater than 50% of the NS-CKD was consumed by 14 of 20 (70%) cats and 12 of 20 cats (60%) consumed >80% of the NS-CKD. The total volume of NS-CKD consumed over the course of the study was statistically greater for the cats classified by owners as having normal appetite (P = 0.046). Increases in body weight were noted for 9 of 14 cats (64.3%) that ingested >50% of the NS-CKD and 1 of 6 cats (16.7%), that ingested ≤ 50% (p = 0.1409) and the group mean % change in body weight was greater in the cats that ingested >50% of the NS-CKD (P = 0.023). The volume of NS-CKD consumed correlated to the % change increases in serum bicarbonate concentration (R = 0.4998;P = 0.02) and was weakly correlated to % change decreases in serum phosphorus concentration (R = 0.0406;P = 0.08). Conclusions: In this pilot study, the NS-CKD was accepted by most cats, no adverse effects were noted, and several findings suggest that the product was associated with ameliorating some metabolic complications which suggest it could be considered in the management of cats with CKD.展开更多

As a mechanism study,only HW significantly inhibited lipid peroxidation in vitro Fenton reaction-mediated ·OH-generating system.Collectively, the results indicate that HW not only effectively eliminated PM from the lungs and blood by enhancing phagocytic activity, but also attenuated the lung injuries by inhibiting lipid peroxidation.

文摘Natural volatile organic compounds(VOCs) extracted from conifers such as P. koraiensis and L. kaempferi have long been studied for their anti-oxidant, anti-proliferative, and anti-inflammatory effects. To evaluate the anti-inflammatory effects of VOCs from P. koraiensis and L. kaempferi, lipopolysaccharide(LPS) was administered to generate a mouse model for inflammation by the nasal route to the lungs and intraperitoneally to the whole body. VOCs of P. koraiensis and L. kaempferi were exposed to the mice by standardized wood panels with closed system. Increased levels of serum IgE and PGE2 were observed after exposure to dexamethasone and VOCs. We further determined the expression levels of inflammatory cytokine mRNA in the LPS-induced inflammation model by the reverse transcription quantitative polymerase chain reaction. Furthermore, the levels of cyclooxygenase-2, tumor necrosis factor-α, interleukin-1β, and interleukin-13 were determined in peripheral blood mononuclear cells. Those inflammatory cytokines and the key enzyme for inflammation cyclooxygenase-2 expression in PBMCs were strongly reversed by dexamethasone and VOCs. Lung tissues after nasal LPS exposure showed increased cytokine mRNA expressions which were suppressed by treatment with dexamethasone and VOCs. Furthermore, the damage induced by LPS was attenuated by dexamethasone and VOCs. In conclusion, the results from the present study indicate that VOCs of P. koraiensis and L. kaempferi have a therapeutic potential in the treatment or prevention of local and systemic inflammation due to their immunosuppressive effects.
文摘Since estrogenic pollutants and phytoestrogens can cause the disorder of the reproductive system, the effects of a soybean milk product(Vegemil^(?) containing 162 ppm isoflavones) on the feto-neonatal development, including male reproductive function, were investigated. Pregnant rats were fed the soybean milk(5% or 100% in drinking water)from gestational day(GD) 6 to parturition or to post-natal day(PND) 56. Specifically, the rats were divided into 4 groups: the control group(drinking water), the GD5% group(5% soybean milk during only the GD period), the GDPND5% group(5% soybean milk during the GD and PND periods), and the GD-PND100% group(100% soybean milk instead of water during the GD and PND periods). During the gestational, lactational, and developmental periods, the reproductive and developmental parameters of dams and offspring were observed. Feeding soybean milk did not affect the birth and physical development of both male and female offspring. At PND57, the weights of the testes and epididymides of F1 males significantly increased by feeding a high concentration of the soybean milk(GDPND100%). In addition, feeding of the soybean milk during both the GD and PND periods(GD-PND5% and GDPND100%) enhanced the sperm counts and motility. The results indicate that soybean milk is safe for embryos,fetuses, and offspring, and improves the post-generational development of male reproductive function.
文摘Purpose: To determine if cats with chronic kidney disease (CKD) would willingly consume an oral nutritional supplement formula (NS-CKD) and to assess associated effects on select clinical and biochemical parameters. Methods: Client-owned cats with CKD classified as International Renal Interest Society (IRIS) stage 2 (n = 7), IRIS stage 3 (n = 12), or IRIS stage 4 (1 cat) were classified by the owners as having normal or variable appetites. The cats were offered 30 ml NS-CKD for 14 days concurrently with a meal in a separate bowl and the amount of the NS-CKD consumed daily was recorded. Bodyweight, physical examination, and serum biochemical profiles were assessed on Days 0, 7, and 14. Results: Greater than 50% of the NS-CKD was consumed by 14 of 20 (70%) cats and 12 of 20 cats (60%) consumed >80% of the NS-CKD. The total volume of NS-CKD consumed over the course of the study was statistically greater for the cats classified by owners as having normal appetite (P = 0.046). Increases in body weight were noted for 9 of 14 cats (64.3%) that ingested >50% of the NS-CKD and 1 of 6 cats (16.7%), that ingested ≤ 50% (p = 0.1409) and the group mean % change in body weight was greater in the cats that ingested >50% of the NS-CKD (P = 0.023). The volume of NS-CKD consumed correlated to the % change increases in serum bicarbonate concentration (R = 0.4998;P = 0.02) and was weakly correlated to % change decreases in serum phosphorus concentration (R = 0.0406;P = 0.08). Conclusions: In this pilot study, the NS-CKD was accepted by most cats, no adverse effects were noted, and several findings suggest that the product was associated with ameliorating some metabolic complications which suggest it could be considered in the management of cats with CKD.

文摘Transitional cell carcinoma(TCC)is the most common tumor of urinary bladder(UB)in dogs.TCC usually has high invasiveness and frequent involvement of trigone region,and therefore most TCC in dogs are judged as unresectable when diagnosed.Moreover,tumor involvement of trigone region and urethra could cause partial or complete urinary tract obstruction.As TCCs are clinically highly malignant,median survival time of the patients with TCCs at any TMN stage has been reported to be less than 1 year regardless of the type of surgical intervention and chemotherapeutic regimen in most previously studies.
