AIM:To investigate the distribution and expressionof C-type natriuretic peptide(CNP)/natriuretic peptide receptor B(NPR-B) in the rectum of a rodent depression model and the interventional effect of Xiaoyaosan(XYS).ME...AIM:To investigate the distribution and expressionof C-type natriuretic peptide(CNP)/natriuretic peptide receptor B(NPR-B) in the rectum of a rodent depression model and the interventional effect of Xiaoyaosan(XYS).METHODS:Male rats(n = 45) of clean grade(200 ± 20 g) were divided into five groups after one week of adaptive feeding:primary control,depression model,low dose XYS,middle dose XYS,and high dose XYS.The animal experiment continued for 3 wk.Primary controls were fed normally ad libitum.The rats of all other groups were raised in solitary and exposed to classic chronic mild unpredictable stimulation each day.XYS groups were perfused intragastrically with low dose,middle dose,and high dose XYS one hour before stimulation.Primary control and depression model groups were perfused intragastrically with normal saline under similar conditions as the XYS groups.Three weeks later,all rats were sacrificed,and the expression levels of CNP and NPR-B in rectum tissues were analyzed by immunohistochemistry,real-time polymerase chain reaction,and Western blotting.RESULTS:CNP and NPR-B were both expressed in the rectum tissues of all rats.However,the expression levels of CNP and NPR-B at both gene and protein levels in the depression model group were significantly higher when compared to the primary control group(n = 9; P < 0.01).XYS intervention markedly inhibited the expression levels of CNP and NPR-B in depressed rats.The expression levels of CNP and NPR-B in the high dose XYS group did not significantly differ from the expression levels in the primary control group.Additionally,the high and middle dose XYS groups(but not the low dose group) significantly exhibited lower CNP and NPR-B expression levels in the rectum tissues of the respectively treated rats compared to the untreated depression model cohort(n = 9; P < 0.01).CONCLUSION:The CNP/NPR-B pathway is upregulated in the rectum of depressed rats and may be one mechanism for depression-associated digestive disorders.XYS antagonizes this pathway at least partially.展开更多
Objective:To explore the optimal scheme of Shengmai Injection combined with other drugs for clinical doctors.Methods:Based on the large-scale data warehouse established by the institute of Clinical Basic Medicine,Chin...Objective:To explore the optimal scheme of Shengmai Injection combined with other drugs for clinical doctors.Methods:Based on the large-scale data warehouse established by the institute of Clinical Basic Medicine,Chinese Academy of Traditional Chinese Medicine,the hospital information system of 22 large-scale tertiary hospitals in China was collected and 1751 patients with angina pectoris who were treated with Shengmai Injection were selected.Louvain algorithm and complex network analysis are used to build the model to summarize the rule of Shengmai Injection in the treatment of angina pectoris of coronary heart disease.Results:On the basis of Shengmai Injection,according to the symptom treatment of angina pectoris of coronary heart disease,nitrates or Suxiao Jiuxin Pill was used to dilate coronary artery;for the treatment of risk factors of angina pectoris,a combination of"insulin+acarbose+bisoprolol+nifedipine+captopril+estazolam"was used to control blood glucose and blood pressure;"Ganmao Qingre Granule+levofloxacin"to treat upper respiratory tract infection and other medication regimens were used as well.For the complications of angina pectoris of coronary heart disease,"furosemide+spironolactone+potassium chloride+magnesium sulfate"was used to reduce edema and“isosorbide nitrate+metoprolol+Shenmai injection+Wenxin Granule"was used to improve symptom of heart failure of qi and yin deficiency type.Conclusion:The therapeutic regimen of Shengmai Injection combined with traditional chinese medicine and western medicine is basically consistent with the current guidelines,but more clinical studies are still needed to explore more effective combination therapy.展开更多

Objective:To observe the curative effect of the complementary and alternative therapies in relieving insomnia for cancer patients.Method:According to the principle of randomization,with meeting the inclusion criteria,...Objective:To observe the curative effect of the complementary and alternative therapies in relieving insomnia for cancer patients.Method:According to the principle of randomization,with meeting the inclusion criteria,the 60 patients who were diagnosed as insomnia caused by liver qi stagnation were randomly divided into the control group and the intervention group.The intervention group(30 individuals treated with mini-needle therapy combined with TCM five elements music),the control group(30 individuals who were treated with single mini-needle therapy),the body buried needle 24 hours/time,5 times for a course of treatment,interval of 2 days between 2 courses,a total of 8 needles embedded.Pittsburgh sleep quality index(PSQI)scores of 2 groups were analyzed using SPSS19.0 statistical software.Results:The total effective rate was 86.1%in the intervention group,and 75.9%in the control group,the difference was statistically significant between 2 groups(P<0.05).The comparison of the PSQI score between the 2 groups propost treatment showed that there were statistically significant differences(P<0.05).The reduction rate of PSQI in the intervention group was 31.7%,indicating that 31.7%was effective,while 23.1%in the control group was ineffective.Conclusion:The mini-needle therapy combined with TCM five elements music could relieve insomnia caused by qi stagnation of liver in patients with advanced cancer.展开更多

OBJECTIVE:To characterize naringenin(NAR) population pharmacokinetics(PPK) in Chinese women with primary osteoporosis.METHODS:Ninety-eight female patients with primary osteoporosis from the Jingshan,Beixinqiao,Jiaodao...OBJECTIVE:To characterize naringenin(NAR) population pharmacokinetics(PPK) in Chinese women with primary osteoporosis.METHODS:Ninety-eight female patients with primary osteoporosis from the Jingshan,Beixinqiao,Jiaodaokou,Chaoyangmen,and Donghuamen communities in Beijing,China,aged 40 to 80 years,received oral Qianggu capsules(250 mg).Blood samples were collected before and at 0.5,1,2,3,4,6,8,10,12,and 24 h after administration.The concentration of NAR in the blood samples was measured using high performance liquid chromatography-tandem mass spectrometry.PPK analyses were performed with nonlinear mixed-effect modeling software(version 7.1.2,PsN3.2.12).The clearance(C1),central distribution volume(V),absorption rate constant(Ka1),peripheral distribution volume(VII),and inter-compartmental clearance(CLII) were set as parameters and estimated by the base model,covariate model,and final model.Kidney-Yang deficiency[Shenyangxu(SYAX)]and liver-kidney-Yin deficiency(Ganshenyinxu) are patterns of symptoms in Traditional Chinese Medicine that were set as covariates,along with age,height,blood urea nitrogen,serum creatinine,alanine transaminase,aspartate transaminase,and hyperlipidemia.Both stepwise forward and backward procedures were accomplished to build models.The final model was evaluated by internal and external validation,visual predictive check,bootstrap,and leverage analysis.RESULTS:A one compartment open model with first order degradation was the best fitted to the concentration-time profiles following oral administration of NAR.The mean of population parameters of the final model,C1,SYAX on C1,V,Ka1,CLII,and VII,were measured to be 37.6 L/h,0.427 L,123 L/h,0.12/h,0.3056,and 1.446,respectively.Inter-individual variability was estimated and SYAX was identified as a significant covariate.CONCLUSION:The population pharmacokinetic model described in this study could effectively characterize the pharmacokinetic profile of NAR following administration of a single dose of oral Qianggu capsules in Chinese women with primary osteoporosis.Among the tested covariates,only SYAX was a significant factor.展开更多

METHODS: Twenty-five subjects...OBJECTIVE: To explore the effect of antithymocyte globulin (ATG)/antilymphocyte globulin (ALG) plus kidney-nourishing Chinese medicinal (KNCM) on se- vere aplastic anemia (SAA). METHODS: Twenty-five subjects of severe aplastic anemia were treated with ATG/ALD plus KNCM be- tween 1992 and 2009, and the clinical data before and after treatment were collected and analyzed. RESULTS: Of the 25 patients, 9 were nearly cured, 6 were improved, 5 were in remission, and 5 failed. The overall effective rate was 80.0%. The 3-year, 5-year, 10-year, 15-year survival rate were respec- tively 98.6%, 97.3%, 97.3%, 67.5%, and median sur- vival time was 180 months. Compared to the condi- tions before administering the medication of ATG/ ALG plus KNCM, after 2 weeks, reticulocyte was first improved (P=0.001), one month later, followed by palette (P=0.037), two months later, by neutrophil cell in peripheral blood (P=0.001); three months lat- er, then by the hemoglobin (P=0.012). By conduct- ing 1-year follow-up, 1 case of complication--parox-ysmal nocturnal hemoglobinuria (PNH) was identi- fied and the patient still alive today. 文摘OBJECTIVE:To characterize naringenin(NAR) population pharmacokinetics(PPK) in Chinese women with primary osteoporosis.METHODS:Ninety-eight female patients with primary osteoporosis from the Jingshan,Beixinqiao,Jiaodaokou,Chaoyangmen,and Donghuamen communities in Beijing,China,aged 40 to 80 years,received oral Qianggu capsules(250 mg).Blood samples were collected before and at 0.5,1,2,3,4,6,8,10,12,and 24 h after administration.The concentration of NAR in the blood samples was measured using high performance liquid chromatography-tandem mass spectrometry.PPK analyses were performed with nonlinear mixed-effect modeling software(version 7.1.2,PsN3.2.12).The clearance(C1),central distribution volume(V),absorption rate constant(Ka1),peripheral distribution volume(VII),and inter-compartmental clearance(CLII) were set as parameters and estimated by the base model,covariate model,and final model.Kidney-Yang deficiency[Shenyangxu(SYAX)]and liver-kidney-Yin deficiency(Ganshenyinxu) are patterns of symptoms in Traditional Chinese Medicine that were set as covariates,along with age,height,blood urea nitrogen,serum creatinine,alanine transaminase,aspartate transaminase,and hyperlipidemia.Both stepwise forward and backward procedures were accomplished to build models.The final model was evaluated by internal and external validation,visual predictive check,bootstrap,and leverage analysis.RESULTS:A one compartment open model with first order degradation was the best fitted to the concentration-time profiles following oral administration of NAR.The mean of population parameters of the final model,C1,SYAX on C1,V,Ka1,CLII,and VII,were measured to be 37.6 L/h,0.427 L,123 L/h,0.12/h,0.3056,and 1.446,respectively.Inter-individual variability was estimated and SYAX was identified as a significant covariate.CONCLUSION:The population pharmacokinetic model described in this study could effectively characterize the pharmacokinetic profile of NAR following administration of a single dose of oral Qianggu capsules in Chinese women with primary osteoporosis.Among the tested covariates,only SYAX was a significant factor.
文摘OBJECTIVE: To explore the effect of antithymocyte globulin (ATG)/antilymphocyte globulin (ALG) plus kidney-nourishing Chinese medicinal (KNCM) on se- vere aplastic anemia (SAA). METHODS: Twenty-five subjects of severe aplastic anemia were treated with ATG/ALD plus KNCM be- tween 1992 and 2009, and the clinical data before and after treatment were collected and analyzed. RESULTS: Of the 25 patients, 9 were nearly cured, 6 were improved, 5 were in remission, and 5 failed. The overall effective rate was 80.0%. The 3-year, 5-year, 10-year, 15-year survival rate were respec- tively 98.6%, 97.3%, 97.3%, 67.5%, and median sur- vival time was 180 months. Compared to the condi- tions before administering the medication of ATG/ ALG plus KNCM, after 2 weeks, reticulocyte was first improved (P=0.001), one month later, followed by palette (P=0.037), two months later, by neutrophil cell in peripheral blood (P=0.001); three months lat- er, then by the hemoglobin (P=0.012). By conduct- ing 1-year follow-up, 1 case of complication--parox-ysmal nocturnal hemoglobinuria (PNH) was identi- fied and the patient still alive today.
